摘要
本文通过10只兔和13例人骨髓细胞4℃保存,证明TC199对骨髓细胞4℃保存是有效的。EDTA·Na2对骨髓细胞的抗凝能力比枸橼酸钠强,肝素抗凝维持时间短,容易产生凝结。TC199-EDTA·Ha2(TCE)保存液与等量骨髓细胞混悬后,在4℃保存3天,骨髓细胞的台盼蓝和伊红拒染细胞的百分率和有核细胞回收率均在70%以上,骨髓粒系祖细胞(Granulocyte-Macrophage Colony-Forming Unit In Culture,简称CFU-C)的回收率为80%,保存5天时骨髓细胞中的粒细胞将出现固缩等退行性变化。临床资料初步表明,4℃保存3天的TCE骨髓细胞的输注是安全的,未见异常反应,可以加速放疗和化疗后骨髓及血液学的恢复。
Ten rabbits and thirteen human bone marrow cells specimens were preserved at 4℃in TC199-EDTA.Na2(TCE),TC 199-heparin and TC 199-citrate-adenine-glucose solutions respectively.The results showed that the anticoagulative effect of EDTA.Na2 on bone marrow cells was greater than that of citrate or heparin.When equal volume of TCE preservation solution was added to bone marrow fluid and preserved at 4℃for 3 days,there was no sign of coagulation and both the viability and recovery rates of bone marrow cells were greater than 70%.The recovery rate of granulocyte-macrophage colony-forming units in culture was approximately 80%.However,after 5 days of storage,pyknotic degeneration of the eells appeared.TCE solution has been used clinically in 5 cases.Autoreinfusion of TCE bone marrow cells preserved for 3 days at 4℃was safe and improved the hematopoietic function Of the patients after chemotherapy in the 3-day period.No adverse effect was observed.
