15甲基前列腺素F_(2α)对离体早孕小鼠的子宫收缩作用

摘要 前列腺素及其类似物在临床的引产和流产效果,与其子宫收缩作用密切有关。早期的药理研究表明,前列腺素的子宫作用与体内激素状态有密切关系,而与已知神经递质的释放及其受体无关[1]。 The interactions between 15-methyl prostaglandin F_(2α)(15-methyl PGF_(2α))and some related agents were studied on early pregna5nt mice uterus in vitro.Results showed that 15-methyl PGF_(2α)could significantly antagonize the inhibitory action induced by aminophylline and isoproterenol on electrically stimulated uterus.The addition of an anti-inflammatory analgesic,indomethacin,at a lower concentration produced 76 percent depression of the amplitude of electrically stimulated contraction.15-Methyl prostaglandin F_(2α)could reverse this inhibitory action of indomethacin.However,when the uterus contraction was totally inhibited by indomethaoin at higher concentration(500μg per milliliter),15-methyl PGF_(2α)was then unable to reveal any antagonistic action even at high concentration.There might be two possibilities,namely the effect of 15-methyl PGF_(2α)upon uterus contraction might be mediated by the endogeneous prostaglandins,or large dosage of indomethacin might impair the basic mechanism of uterine contraction,so that 15-methyl PGF_(2α)could no longer exert any stimulatory effect on the myometrium.15-methyl PGF_(2α)could exhibit its stimulatory action on spontaneous contraction of the uteri in Kreb-Henseleit solution with different concentration of calcium.
