基于BDNF Val66Met单核苷酸多态性分析的针刺治疗偏头痛的个体疗效差异研究

Difference in Individual Efficacy of Acupuncture for Migraine Based on BDNF Val66Met Single Nucleotide Polymorphism Analysis

目的:探索脑源性神经营养因子(BDNF)Val66Met基因单核苷酸多态性对月经性偏头痛受试者针刺疗效差异的影响。方法:对纳入的70例月经性偏头痛受试者进行统一针刺治疗,4周后以治疗前后头痛天数的缓解率大于等于50%作为针刺“有效”的疗效评判标准筛选针刺疗效差异受试者,分为针刺有效组与针刺无效组2组。采集受试者全血,用实时荧光定量PCR TapMan探针法检测BDNF Val66Met基因,比较疗效差异受试者基因型和等位基因分布的差异性。结果:针刺有效组BDNF基因型分布为VAL/VAL15%,VAL/MET62%,MET/MET23%;无效组为VAL/VAL26%,VAL/MET52%,MET/MET22%。针刺有效组BDNF等位基因分布为VAL46%,MET54%;无效组为VAL52%,MET48%。两组BDNF Val66Met基因型及等位基因分布的差异均无统计学意义(P>0.05)。结论:尚未发现BDNF Val66Met单核苷酸多态性与月经性偏头痛针刺疗效差异的相关性。

Objective:To explore the impact of single nucleotide polymorphism(SNP)on different acupuncture responders with menstrual migraine.Methods:Seventy participants received 5 times acupuncture treatment weekly for 4 weeks.Then they were allocated into an acupuncture positive group(PG)and a negative group(NG)according to evaluation result(those whose remission rate of headache days was not less than 50%were positive,and the others were negative).After that,the BDNF Val66 Met gene of the participants were detected to explore the SNP impact on different acupuncture responders.Results:A total of 70 patients were included in the trial,and 67 patients,of which 26 were effective and 41 were ineffective,finished the study.After analysis,we get the following results:the BDNF genotype distribution of PG was VAL/VAL 15%,VAL/MET 62%,MET/MET 23%;the BDNF genotype distribution of NG was VAL/VAL 26%,VAL/MET 52%,MET/MET 22%.The distribution of BDNF alleles in PG was VAL 46%and MET 54%;the distribution of BDNF alleles in NG was VAL 52%and MET 48%.There was no significant difference in the frequency of BDNF Val66 Met genotypes and alleles between patients with menstrual migraine who have different efficacy of acupuncture treatment(P>0.05).Conclusion:No correlation between the BDNF Val66 Met SNP and the difference in the efficacy of acupuncture for menstrual migraine had been found.
