摘要
目的:探讨土茯苓有效成分落新妇苷对大鼠血管平滑肌细胞(VSMCs)的增殖、迁移及表型转换的影响及其可能的机制。方法:采用CCK-8法检测不同浓度(10、25、50μmol/L)落新妇苷对血小板衍生因子-BB(PDGFBB)(20ng/ml)诱导的VSMCs增殖的影响;划痕实验用于检测落新妇苷对PDGF-BB(20ng/ml)诱导的VSMCs迁移能力影响;免疫印迹法检测落新妇苷作用后的PCNA、α-SMA、OPN、AMPK及其磷酸化蛋白表达水平;流式细胞术检测落新妇苷作用后对VSMCs凋亡的影响。结果:与对照组(NC)相比,落新妇苷显著抑制了PDGF-BB诱导的VSMCs增殖(^(∗)P<0.05,^(∗∗)P<0.01)、迁移、表型转换。此外,落新妇苷激活了腺苷酸活化蛋白激酶(AMPK)信号。结论:我们首次发现落新妇苷可以显著抑制PDGF-BB诱导的VSMCs过度增殖和迁移,机制上,其可能与AMPK的激活有关。该研究为临床上血管介入术后再狭窄的防治提供了参考。
Objective:To discuss the effect of Astilbin on the proliferation,migration and phenotypic transformation for vascular smooth muscle cells(VSMCs)in rats and its possible mechanisms.Methods:CCK-8 was used to detect the effect of different concentrations(10,25,50μmol/L)of lassinoside on the proliferation of VSMCs induced by platelet derived growth factor-BB(PDGF-BB)(20 ng/ml).The scratch test was used to detect the effect of Astilbin on the migration of VSMCs induced by PDGF-BB(20 ng/ml).Western blotting was used to detect the expression levels of PCNA,α-SMA,OPN,AMPK and their phosphorylated proteins.Flow cytometry was used to detect the effect of astilbin on the apoptosis of VSMCs.Results:Astilbin significantly inhibited the proliferation,migration and phenotypic transformation of PDGF-BB-induced VSMCs(^(∗)P<0.05,^(∗∗)P<0.01)compared with the NC group.In addition,Astilbin activates adenosine activated protein kinase(AMPK)signaling.Conclusion:For the first time,we found that Astilbin significantly inhibited the over-proliferation and migration of PDGF-BB-induced VSMCs,which may be related to AMPK activation.This study provides a reference for the prevention and treatment of restenosis after vascular intervention.
