摘要
目的:研究芪蛭真武汤对慢性心力衰竭大鼠心肌组织转化生长因子-β(TGF-β)、脑钠肽(BNP)、α-平滑肌肌动蛋白(α-SMA)、白细胞介素-6(IL-6)表达的影响。方法:将28只Dahl盐敏高血压大鼠随机分为模型组、卡托普利组、芪蛭真武汤低剂量组、芪蛭真武汤高剂量组,空白组设置7只SS-13BN盐不敏大鼠。模型组及各治疗组均予8%高盐饲料喂养4周造模,空白组SS-13BN盐不敏大鼠予8%高盐饲料喂养。造模成功后模型组及空白组予无菌蒸馏水10ml·kg^(-1),卡托普利组予以卡托普利片0.5g·kg^(-1),芪蛭真武汤低剂量组予芪蛭真武汤11.1g·kg^(-1),芪蛭真武汤高剂量组予芪蛭真武汤22.2g·kg^(-1)。连续干预治疗8周。ELISA检测TGF-β、BNP水平,HE、Masson染色观察心肌组织病理学变化。蛋白免疫印迹法(Western blot)检测α-SMA、IL-6蛋白表达。结果:与空白组比较,模型组TGF-β、BNP水平明显升高,模型组α-SMA、IL-6表达均升高,差异有统计学意义(P<0.01);与模型组比较,各治疗组TGF-β、BNP水平均下降(P<0.05),其中各中药组TGF-β、BNP水平与α-SMA、IL-6表达均显著下降,差异有统计学意义(P<0.01)。结论:芪蛭真武汤可以降低慢性心力衰竭大鼠心肌组织TGF-β、α-SMA水平,抑制心肌纤维化,阻止心室重构的发生;减少IL-6的释放,抑制BNP的分泌,减少炎性细胞因子的释放,减轻心肌组织炎症反应,从而改善心功能。
Objective:To study the effect of Qizhi Zhenwu Decoction on the expression of transforming growth factor-β(TGF-β),Brain Natriuretic Peptide(BNP),α-smooth muscle actin(α-SMA)and interleukin-6(IL-6)in myocardium for rats with chronic heart failure.Methods:28 Dahl salt-sensitive hypertensive rats were randonly divided into a model group,a captopril group,a Qizhi Zhenwu Decoction low dose group and a Qizhi Zhenwu Decoction high dose group.7 SS-13 BN salt-insensitive rats were set up in the blank group.The model group and the each treatment group were fed with 8%high-salt diet for 4 weeks,and the SS-13 BN salt-insensitive rats in the blank group were fed with 8%high-salt diet.After successful modeling,the model group and the blank group were treated with aseptic distilled water 10 ml.kg^(-1),the captopril group with 0.5 g.kg^(-1),the Qizhi Zhenwu Decoction low dose group were given Qizhi Zhenwu Decoction 11.1 g.kg^(-1),the Qizhi Zhenwu Decoction high dose group were given Qizhi Zhenwu Decoction 22.2 g.kg^(-1).Continuously intervene and treat 8 weeks.The levels of TGF-βand BNP were detected by ELISA,and the pathological changes of myocardial tissue were observed by HE and Masson staining.The protein expressions ofα-SMA and IL-6 were detected by Western blotting(Western blot).Results:Compared with the blank group,the levels of TGF-βand BNP in the model group increased signigicantly,and the expression ofα-SMA and IL-6 in the model group increased significantly,the difference was statistically significant(P<0.01);compared with the model group,the levels of TGF-βand BNP in the each treatment group were decreased,the difference was statistically significant(P<0.05),and the levels of TGF-βand BNP and the expression ofα-SMA and IL-6 in the each traditional Chinese medicine group was significantly lower than those in the model group(P<0.01).Conclusion:Qizhi Zhenwu Decoction can reduce the levels of TGF-βandα-SMA in myocardial tissue of rats with chronic heart failure,inhibit myocardial fibrosis,prevent the occurence of ventricular remodeling,reduce the release of IL-6,inhibit the secretion of BNP,reduce the release of myocardial tissue,and improve cardiac function.
