辛前甘桔汤对慢性鼻-鼻窦炎模型大鼠炎症因子表达及p65、p-P38蛋白的研究 被引量:1

摘要 目的:探讨辛前甘桔汤对慢性鼻-鼻窦炎模型大鼠核因子κ-B(NF-κB)p65、p38丝裂素活化蛋白激酶(p38-MARK)及相关炎症因子水平的影响。方法:健康SD大鼠随机分为空白对照组、慢性鼻-鼻窦炎模型组、克拉霉素组和辛前甘桔汤组。除空白对照组大鼠外,建立模型,给予相应药物干预。采用ELISA法分别检测血清及鼻黏膜肿瘤坏死因子α(TNF-α)、干扰素γ(IFN-γ)、白细胞介素-1β(IL-1β)的表达,Real-time RT-PCR法检测鼻黏膜黏蛋白5AC(MUC5AC),Western-Blot法检测NF-κB、p38-MARK及磷酸化指标。结果:慢性鼻-鼻窦炎模型组与空白对照组比较,血清及鼻黏膜TNF-α、IL-1β、MUC5AC的水平均显著升高(P<0.001),IFN-γ水平显著降低(P<0.001),p-NF-κB、p-P38含量明显升高(P<0.01)。与西药组比较,辛前甘桔汤组血清TNF-α水平有差异(P<0.05),IL-1β、IFN-γ水平有显著差异(P<0.001),鼻黏膜TNF-α、IFN-γ水平有差异(P<0.05),IL-1β水平有显著差异(P<0.001),MUC5AC水平无差异(P>0.05),p-NF-κB、p-P38表达无明显差异(P>0.05)。结论:辛前甘桔汤可以降低鼻黏膜内p65、p-P38蛋白含量,进而影响鼻黏膜内炎症因子TNF-α、IL-1β、IFN-γ分泌,同时抑制黏蛋白分泌;辛前甘桔汤可能是通过调节NF-κB、p38 MARK信号通路,缓解慢性鼻-鼻窦炎模型大鼠的黏膜炎症及高分泌状态。 Objective: To investigate the effect of Xinqian Ganjie Decoction on the levels of nuclear factor-κB p65, p38 mitogen activated protein kinase(p38 mark) and related inflammatory factors for rats with chronic rhinosinusitis. Methods: Healthy SD rats were randomly divided into a blank controlled group, a chronic rhinosinusitis model group, a clarithromycin group and a XinqianGanjie Decoction group. In addition to the blank controlled group, the model was established and the corresponding drug intervention was given. The expression of tumor necrosis factor-α(TNF-α), interferon-γ(IFN-γ) and interleukin-1β(IL-1β) in serum and nasal mucosa were detected by ELISA, MUC5 AC(MUC5 AC) in nasal mucosa was detected by real-time RT-PCR, NF-κB, p38 mark and phosphorylation index were detected by Western blot. Results: Compared with the blank controlled group, the levels of TNF-α, IL-1β and MUC5 AC in serum and nasal mucosa of the chronic rhinosinusitis model group were significantly increased(P<0.001), the levels of IFN-γ were significantly decreased(P<0.001), and the contents of p-nf-κB and p-p38 were significantly increased(P<0.01). Compared with the western medicine group, the levels of serum TNF-α, IL-1β and IFN-γ, nasal mucosa TNF-α and IFN-γ, IL-1β, MUC5 AC, p-nf-κ B and p-p38 in the XinqianGanjie Decoction group were significantly different(P<0.05), P<0.05, P<0.001, P>0.05, (P<0.05). Conclusion: Xinqian Ganjie decoction can reduce p65 and p-P38 protein content in nasal mucosa, and then affect the secretion of inflammatory factors TNF-α, IL-1β and IFN-γ in nasal mucosa, and inhibit the secretion of mucin. Conclusion: Xinqian Ganjie Decoction may alleviate the mucositis and hypersecretion of chronic rhinosinusitis model rats by regulating NF-κ B and p38 mark signaling pathway.
