摘要
目的探讨含钾通道四聚化结构域15(KCTD15)基因在人群脂肪组织中表达水平与临床指标的相关性,分析KCTD15的潜在作用及机制。方法收集2022年6-9月陆军军医大学第二附属医院骨科及普通外科39例手术患者的脂肪组织,根据体质指数(BMI)将受试者分为超重肥胖组(n=22)和正常体重组(n=17)两组。采用RT-PCR法测定两组共39例患者脂肪组织中KCTD15 mRNA表达,以甘油醛-3-磷酸脱氢酶(GAPDH)的A值作为内参照,计算KCTD15/GAPDH的相对表达量,分析KCTD15表达量与临床指标的相关性。结果超重肥胖组BMI、腰围、收缩压、舒张压、肌酐均高于正常体重组(t=13.059、9.781、2.185、2.323、2.148),血清钾离子浓度明显低于正常体重组(t=-4.582),差异均有统计学意义(P均<0.05);超重肥胖组脂肪组织中KCTD15 mRNA水平明显低于正常体重组,差异有统计学意义(P<0.01)。KCTD15 mRNA水平与BMI、腰围、丙氨酸氨基转移酶、总胆红素、直接胆红素成负相关(r=-0.417、-0.468、-0.398、-0.396、-0.420,P均<0.05),与血清钾离子浓度成正相关(r=0.401,P<0.05)。结论脂肪组织中的KCTD15 mRNA的表达量与BMI、腰围、胆红素成负相关,与血清钾离子成正相关,提示KCTD15基因可能在肥胖的病理生理机制中发挥一定作用。
Objective To explore the role and underlying mechanism of potassium channel tetramerization domain containing 15(KCTD15)gene by analyzing the correlation between the expression level of KCTD15 gene in human adipose tissue and clinical indicators.Methods The adipose tissue samples of thirty-nine patients undergoing orthopedic and general surgery in the Army Medical University from June to September 2022 were collected.According to body mass index(BMI),the subjects were divided into overweight and obese group(n=22)and normal weight group(n=17).The levels of KCTD15 mRNA in thirty-nine patients were detected by reverse transcription PCR(RT-PCR).The correlation between KCTD15 expression level and clinical indicators was analyzed.Results Compare to the normal weight group,BMI,waist circumference,systolic blood pressure,diastolic blood pressure and creatinine were higher in overweight and obese group(t=13.059,9.781,2.185,2.323,2.148),while serum potassium concentration in overweight and obese group was significantly lower(t=-4.582),all the differences were statistically significant(all P<0.05).The KCTD15 mRNA level of adipose tissue was significantly lower in the overweight and obese group than that in normal weight group(P<0.01).Correlation analysis showed that the KCTD15 mRNA expression in all patients were negatively correlated with BMI,waist circumference,alanine aminotransferase,total bilirubin and direct bilirubin(r=-0.417,-0.468,-0.398,-0.396,-0.420;all P<0.05),and was positively correlated with serum potassium concentration(r=0.401,P<0.05).Conclusion The KCTD15 mRNA level of adipose tissue was negatively correlated with BMI,waist circumference and bilirubin,and was positively correlated with serum potassium concentration,suggesting that KCTD15 gene might play a crucial role in the pathophysiological mechanism of obesity.
