摘要
钙网蛋白(CRT)是一种主要存在于细胞内质网中高度保守的钙结合蛋白,由N、P和C结构功能区组成,在生物体内分布广泛。寄生虫CRT由虫体分泌或在虫体表面表达,在虫体感染与免疫过程中发挥重要作用。寄生虫感染宿主后,CRT作为一种抗原成分被宿主免疫系统识别,通过促进细胞成熟增殖分化,分泌细胞因子等方式提供免疫保护;将重组CRT作为疫苗免疫小鼠,在攻毒的小鼠模型中提供免疫保护。同时,寄生虫CRT通过干扰宿主补体激活的初始阶段或作为毒力因子参与寄生的建立及维持。不仅如此,CRT还可通过免疫佐剂、抑制血管生长与受体结合等方式发挥抑癌作用。本文将围绕上述内容展开综述,以期为探索寄生虫感染防治新途径提供新思路。
Calreticulin(CRT)is a highly conserved calcium-binding protein which mainly exists in the endoplasmic reticulum.It is composed of the structural and functional domain of N,P and C and is widely distributed in organisms.Parasite CRT is secreted or expressed on the surface of parasite,which plays an important role in the process of infection and immunity.After parasites infect the host,CRT is recognized by the host immune system as an antigen component,which provides immune protection by promoting cell maturation,proliferation and secreting cytokines.Recombinant CRT is used as a vaccine to immunize mice to provide immune protection in the challenged mouse model.At the same time,parasite CRT participates in the establishment and maintenance of parasitism by interfering with the initial stage of host complement activation or as a virulence factor.Moreover,CRT can also inhibit cancer by means of immune adjuvants,inhibiting angiogenesis and binding to receptors.This article will review the above contents in order to provide new ideas for exploring new ways of prevention and treatment of parasite infection.
