江门市公共场所淋浴水中嗜肺军团菌污染分子溯源研究

Molecular traceability of Legionella pneumophila contamination in shower water of public places in Jiangmen city

目的通过分析江门市公共场所淋浴水中分离的嗜肺军团菌的基因特征,初步建立嗜肺军团菌分子分型数据库,为卫生行政部门实施监督提供科学依据。方法对分离出来的嗜肺军团菌进行血清分型,并使用脉冲场凝胶电泳(PFGE)进行分子分型,经BioNumerics软件进行聚类分析,按照相似系数>85%的不同菌株归为同一聚类,获得克隆系。结果66株嗜肺军团菌有6种血清群,其中Ⅰ型有37株占56.06%,为优势菌型。嗜肺军团菌有38种PFGE型别,带型的相似系数为18.20%~100.00%,菌株之间PFGE带型差异较大。相同的PFGE带型≥3条有6种型别,分别为P8、P9、P11、P21、P25和P26,无优势带型。最大的克隆系是3和4。结论从公共场所淋浴水分离的嗜肺军团菌呈基因多样性,也具有一定的遗传稳定性。

Objective By analyzing the genetic characteristics of Legionella pneumophila isolated from shower water in public places in Jiangmen city,a molecular typing database of Legionella pneumophila was initially established to provide a scientific basis for the implementation of supervision by the health administration department.Methods The isolated Legionella pneumophila strains were serotyped,and the molecular typing was performed by pulsed-field gel electrophoresis(PFGE).Cluster analysis was performed by using BioNumerics software,and different strains were grouped into the same cluster according to the similarity coefficient>85%.Results A total of 66 strains of Legionella pneumophila had 6 serogroups;among which,37 strains were serotypeⅠ,accounting for 56.06%,which was the dominant type.There were 38 PFGE types of Legionella pneumophila,and the similarity coefficient of band types was 18.20%-100.00%.There were great differences in PFGE band types among strains.There were 6 types of the same PFGE band type(P8,P9,P11,P21,P25 and P26)with more than 3 PFGE bands.There was no dominant band type.The largest clone lines were 3 and 4.Conclusion The isolates of Legionella pneumophila from shower water in public places showed genetic diversity and some genetic stability.
