摘要
目的检测新乡市不同环境土壤中弓形虫污染情况,为控制该市土源性弓形虫病的传播奠定基础。方法从2017年10月至2018年5月,共取新乡市不同地区土壤265份,其中高校校园土壤90份、公园土壤85份、居民区土壤90份,提取土壤样本弓形虫DNA,采用PCR技术扩增弓形虫ITS-1基因,然后对PCR产物进行凝胶电泳成像,观察结果并计算阳性率。结果265份土壤样本中,有41份扩增出341 bp大小的目的条带,阳性样品基因测序及BLAST分析显示,获得的目的基因序列与GenBank中弓形虫RH虫株ITS-1基因核苷酸序列(GenBankTM accession number:AY259044.1)的相似性为100%。弓形虫总检出率为15.47%(41/265),其中高校校园、公园、居民区土壤样本中弓形虫阳性率分别为21.11%(19/90)、10.59%(9/85)和14.44%(13/90)。结论新乡市不同环境土壤中弓形虫污染较为普遍。
Objective This study was to investigate the presence of T.gondii in different environmental soil samples in Xinxiang region,China,laying the foundation for controlling the spread of soil-borne toxoplasmosis in this region.Methods From October 2017 to May 2018,a total of 265 soil samples were taken from different sites(90 from universities,85 from parks and 90 from residential areas)located in the Xinxiang region.The DNA from soil samples was extracted.PCR reactions targeting the ITS-1 gene were performed for specific T.gondii detection.Results A clear amplicon corresponding to the expected size of 341 bp was observed in 41 of 265 soil samples.The sequencing and BLAST analysis of the positive samples showed that the nucleotide sequences of the target gene were 100%similar to the ITS-1 sequence of the T.gondii RH strain(GenBankTM accession number AY259044.1).The PCR detection of soil samples showed that,41 samples(15.47%)of 265 samples were found positive for T.gondii.The positive rate of T.gondii DNA in soil samples from universities,parks,residential areas were 21.11%(19/90),10.59%(9/85)and 14.44%(13/90),respectively.Conclusion The contamination of T.gondii indifferent environmental soil samples in Xinxiang region was widespread.
