白色念珠菌对小鼠阴道上皮中β-防御素-2和miR-124表达的影响

Regulation of beta defensin-2 and miR-124 expression in mouse vaginal epithelial cells by Candida albicans

摘要 目的研究白色念珠菌对β-防御素-2和miR-124在小鼠阴道上皮细胞中表达的影响。方法用0、10~1、10~2、10~3个/mL的白色念珠菌作用于小鼠阴道上皮细胞,并于0、24、48、72 h用MTT检测细胞的增长率,用免疫荧光和免疫印迹检测β-防御素-2在小鼠阴道表达的变化,用定量PCR检测miR-124表达量的变化。结果用不同浓度的白色念珠菌作用于小鼠阴道上皮细胞,MTT法检测并计算得到在48 h、102个/mL白色念珠菌作用于小鼠阴道上皮细胞的增长率最低为(23.11±0.21)%。用10~2个/mL白色念珠菌感染小鼠阴道上皮细胞,通过免疫荧光和Western blot检测,发现随着感染时间的增加,β-防御素-2的表达在逐渐增加,且72 h表达量最高。而miR-124表达量逐渐减低为(0.62±0.03)。结论白色念珠菌对小鼠阴道上皮细胞中β-防御素-2的表达与miR-124表达成负相关。 Objective To study the effect of Candida albicans on the expression of beta-defensin-2 and microRNA-124 in mouse vaginal epithelial cells.Methods Vaginal epithelial cells of mice were treated with 0,10~1,10~2 and 10~3/mL Candida albicans.The growth rate of the cells was measured by MTT at 0,24,48 and 72 h.The expression of beta-defensin-2 in the vagina of mice was detected by immunofluorescence and immunoblotting.The expression of microRNA-124 was detected by quantitative PCR.Results Different concentrations of Candida albicans were used in mouse vaginal epithelial cells.MTT method was used to detect and calculate the growth rate of vaginal epithelial cells in mice at 48 hours.The growth rate of mouse vaginal epithelial cells was reduced to(23.11±0.21)%when treated with 102/mL Candida albicans.102/mL Candida albicans were used to infect vaginal epithelial cells of mice.The expression of beta-defensin-2 in 10~2/mL Candida albicans treated vaginal epithelial cells was increased gradually with the increase of infection time,and the highest expression was found at 72 hours;the expression of microRNA-124 decreased gradually to(0.62±0.03).Conclusion The expression of beta-defensin-2 in mouse vaginal epithelial cells infected with 102/mL Candida albicans was negatively correlated with the expression of microRNA-124.
