miRNA-767-3p在结直肠癌中的表达与患者临床病理特征及预后的关系

Relationship between expression of miRNA-767-3p in colorectal cancer and clinicopathological features and prognosis of patients

目的探讨miRNA-767-3p在结直肠癌中的表达情况,并分析其与临床病理特征及预后的关系。方法选取2013年1月至2015年12月于宝鸡市人民医院和渭南市中心医院诊治的82例结直肠癌患者和65例结直肠腺瘤患者作为研究对象,并分别命名为结直肠癌组和结直肠腺瘤组,同时选取同期50名健康体检者作为健康对照组。采用实时荧光定量PCR(qRT-PCR)检测结直肠癌组织、结直肠腺瘤组织、癌旁正常组织和三组血清miRNA-767-3p水平。对比miRNA-767-3p在不同组织和不同人群血清中的差异,分析组织与血清中miRNA-767-3p关系,并分析结直肠癌组织中miRNA-767-3p水平与临床病理特征及其预后的关系。结果miRNA-767-3p的表达由高到低依次为结直肠癌组织、结直肠腺瘤组织和癌旁正常组织,差异有统计学意义(P<0.05);血清miRNA-767-3p水平由高到低依次为结直肠癌组、结直肠腺瘤组和健康对照组,差异有统计学意义(P<0.05);结直肠癌组织与血清中miRNA-767-3p水平呈正相关(r=0.943,P<0.05)。miRNA-767-3p在结直肠癌不同浸润深度、淋巴结转移、远处转移及临床分期上表达差异有统计学意义(P<0.05)。血清miRNA-767-3p高表达者总生存期(57.89%vs.81.82%)和无进展生存期生存率(47.36%vs.72.73%)均低于低表达者,差异有统计学意义(χ2=6.599,P=0.001;χ2=6.775,P=0.009)。结论miRNA-767-3p在结直肠癌患者中高表达,与浸润深度、淋巴结转移、远处转移及临床分期密切相关,在一定程度上有预后价值。

Objective To investigate the relationship between expression of miRNA-767-3 p in colorectal cancer and clinicopathological features and prognosis of patients.Methods 82 patients with colorectal cancer and 65 patients with colorectal adenoma who were treated in our hospital from January 2013 to December 2015 were selected as the research objects.They were named as colorectal cancer group and colorectal adenoma group,respectively.50 healthy people were selected as the healthy control group.The levels of microRNA-767-3 p in the colorectal cancer tissue,colorectal adenoma tissue,adjacent normal tissues and serum were detected by real-time fluorescence quantitative PCR(qRT-PCR).Results The expression levels of microRNA-767-3 p from high to low were colorectal cancer tissues,colorectal adenoma tissues and adjacent normal tissues;the differences were statistically significant(P<0.05).The serum levels of microRNA-767-3 p from high to low were colorectal cancer group,colorectal adenoma group and healthy control group;the differences were statistically significant(P<0.05).The levels of microRNA767-3 p in colorectal cancer tissues and serum were significantly correlated(r=0.943,P<0.05).The expression levels of microRNA767-3 p in different invasion depth,lymph node metastasis,distant metastasis and clinical stage of colorectal cancer were significantly different(P<0.05).The overall survival(57.89%vs.81.82%)and progression-free survival rate(47.36%vs.72.73%)of patients with high expression level of serum microRNA-767-3 p were lower than those with low expression level;the differences were statistically significant(χ2=6.599,P=0.001;χ2=6.775,P=0.009).Conclusion The expression level of microRNA767-3 p in patients with colorectal cancer was closely related to the depth of invasion,lymph node metastasis,distant metastasis and clinical stage,which could be used to predict the survival and prognosis to a certain extent.
