茶多酚、黄芩苷、原花青素拮抗脂多糖影响牙周膜成纤维细胞生理活性的差别研究 被引量:1

摘要 目的分析受脂多糖(LPS)影响的牙周膜成纤维细胞(PDLCs)在茶多酚、黄芩苷、原花青素干预后的效果及差别。方法2018年7月—2019年10月,将256孔PDLCs均分为4组,分别为空白组(滴入9.0%的LPS培养液1.0 ml+0.9%氯化钠溶液1.0 ml),原花青素组(滴入9.0%的LPS培养液1.0 ml+5 mg/ml的原花青素1.0 ml),黄芩苷组(滴入9.0%的LPS培养液1.0 ml+5 mg/ml的黄芩苷1.0 ml),茶多酚组(滴入9.0%的LPS培养液1.0 ml+5 mg/ml的茶多酚1.0 ml),每组均64孔,连续培养7 d。比较四组干预前后PDLCs的细胞凋亡率、增殖活性、前列腺素内氧化酶还原酶(COX-2)和超氧化物歧化酶(SOD)含量、白介素1(IL-1β)和前列腺素E2(PGE2)含量的变化。结果干预后比较,三组(原花青素组、黄芩苷组、茶多酚组)PDLCs的细胞凋亡率、PGE2和IL-1β含量、COX-2含量低于空白组,PDLCs的增殖活性、SOD含量高于空白组(P<0.05)。两两比较,茶多酚组PDLCs的细胞凋亡率低于原花青素组、黄芩苷组,茶多酚组PDLCs的增殖活性高于原花青素组、黄芩苷组,茶多酚组PGE2和IL-1β含量均低于原花青素组和黄芩苷组,茶多酚组COX-2含量低于原花青素组、黄芩苷组,茶多酚组SOD含量高于原花青素组、黄芩苷组(P<0.05)。结论在LPS影响PDLCs生理活性的背景下,应用茶多酚、黄芩苷、原花青素干预均有不同程度效果,其中又以茶多酚效果最佳。 Objective To analyze the effects and differences of periodontal membrane fibroblasts(PDLCs)influenced by lipopolysaccharide(LPS)after the intervention of tea polyphenols,baicalin and procyanidins.Methods From July,2018 to October,2019,256-well PDLCs were divided into four groups:blank group(1.0 ml of 9.0%LPS culture solution+1.0 ml of 0.9%sodium chloride solution)and procyanidin group(1.0 ml of 9.0%LPS culture solution+1.0 ml of 5 mg/ml procyanidin).Baicalin group(9.0%LPS culture solution 1.0 ml+5 mg/ml baicalin 1.0 ml)and tea polyphenols group(9.0%LPS culture solution 1.0 ml+5 mg/ml tea polyphenols 1.0 ml),each with 64 holes,were cultured continuously for 7 days.The apoptosis rate,proliferation activity,contents of Cyclooxygenase-2(COX-2)and Superoxide dismutase(SOD),interleukin-1(IL-1β)and prostaglandin E2(PGE2)of PDLCs before and after intervention were compared among the four groups.Results After intervention,the apoptosis rate of PDLCs,PGE2 and IL-1 content,COX-2 content of the three groups(proanthocyanidin group,baicalin group,and tea polyphenol group)were lower than that of the blank group,and the proliferation activity of PDLCs and SOD content were higher than that of the blank group,and the differences were statistically significant(P<0.05).Compared with procyanidins and baicalin,the apoptosis rate of PDLCs,proliferation activity of PDLCs,contents of PGE2 and IL-1β,COX-2 and SOD in tea polyphenols group were lower than those in procyanidins and baicalin groups(P<0.05).Conclusion Under the background of lipopolysaccharide affecting the physiological activity of periodontal membrane fibroblasts,the application of tea polyphenols,baicalin and procyanidins all had different degrees of effects,among which tea polyphenols had the best effect.
