HPLC法测定莲子心提取物中莲心碱、异莲心碱和甲基莲心碱的含量

Content determination of liensinine, isoliensinine and neferine in the lotus plumule extracts by HPLC

【目的】建立高效液相色谱法测定莲子心提取物中指标成分莲心碱、异莲心碱、甲基莲心碱含量的方法,为莲子心提取物的质量控制提供依据。【方法】Gemini-NX5U C18色谱柱(4.6 mm×250 mm,5μm),乙腈-甲醇-水-三乙胺(40∶20∶40∶0.12)作为流动相,流速1.0 ml/min,检测波长282 nm,柱温30℃。【结果】莲心碱在进样质量0.067~0.670μg,异莲心碱在进样质量0.107~1.070μg,甲基莲心碱在进样质量0.148~1.480μg线性关系良好。方法稳定性、重复性好。莲心碱平均回收率为99.75%、异莲心碱平均回收率为100.53%、甲基莲心碱平均回收率为101.05%;RSD值分别为1.57%、1.54%和1.14%。【结论】所建立的高效液相色谱法简单快速,稳定性高,重复性好,可以用于莲子心提取物的质量控制。

【Objective】To establish a method to determine the contents of liensinine,isoliensinine and neferine in the lotus plumule extracts by HPLC,and provide the basis for quality control of lotus plumule extract.【Methods】The chromatographic separation was performed on a Gemini-NX5U C18 column(4.6 mm×250 mm,5μm)with the elution of acetonitrile-methanol-water-triethylamine(40:20:40:0.12).The flow rate was 1.0 ml/min and the column temperature was 30℃.【Results】The sample mass of liensinine was 0.067-0.670μg,the sample mass of isoliensinine was 0.107-1.070μg and it was 0.148-1.480μg in neferine.All compounds showed satisfactory linearity within the ranges.The method validation showed satisfactory stability and repeatability.The overall recoveries for the three compounds were 99.75,100.53%and 101.05%,respectively.RSD values were 1.57%,1.54%and 1.14%,respectively.【Conclusion】The established HPLC standard is simple and quick,stable,and repeatable,which could ensure the quality control of lotus plumule extracts.
