野蔷薇苷抗大鼠急性低压缺氧脑损伤作用研究 被引量:1

Effect of rosamultin against the brain impairment in acute hypobaric hypoxia-induced rats

摘要 【目的】研究野蔷薇苷对急性低压缺氧诱导的大鼠脑损伤的保护作用,并探讨其可能的机制。【方法】将50只清洁级成年雄性SD(Sprague-Dawley)大鼠(体质量220~250 g),随机分为5组:正常对照组、缺氧模型组、野蔷薇苷高、低剂量组及阳性药(地塞米松)组,每组10只。连续灌胃给药3次后,除正常对照组外,其余各组均运用低压氧舱建立大鼠急性低压缺氧模型;采用生化手段检测大鼠血浆一氧化氮(nitric monoxide,NO)、丙二醛(malondialdehyde,MDA)含量和超氧化物歧化酶(superoxide dismutase,SOD)活性及脑组织中乳酸脱氢酶(lactic dehydrogenase,LDH)活性;利用比重法测定大鼠脑含水量变化;采用免疫组织化学染色法检测大鼠脑组织中水通道蛋白1(aquaporin1,AQP1)和水通道蛋白4(aquaporin4,AQP4)表达情况。【结果】与缺氧模型组相比,野蔷薇苷可显著提高急性低压缺氧大鼠血浆NO含量和SOD活性(P<0.01),降低MDA含量(P<0.01),降低脑组织中LDH活性(P<0.01)和脑含水量(P<0.05),减少AQP1、AQP4阳性细胞表达数量(P<0.01)。【结论】野蔷薇苷可提升大鼠抗氧化能力,有效减轻大鼠脑水肿程度,进而保护急性低压缺氧诱导的大鼠脑损伤。 【Objective】To study the protective effect of rosamultin on the brain impairment in acute hypobaric hypoxia-induced rats,and explore its underlying mechanism.【Methods】A total of 50 clean-grade adult male Sprague-Dawley(SD)rats(body mass 220-250 g)were randomly divided into 5 groups:control group,hypoxic model group,high-dose rosamultin intervention group,low-dose rosamultin intervention group and dexamethasone group,10 rats per group.Except for the control group,the models of acute hypobaric hypoxia were established in the other groups by using hypobaric oxygen cabin.The contents of nitric monoxide(NO),malondialdehyde(MDA)and superoxide dismutase(SOD)activity in plasma,as well as the activity of lactic dehydrogenase(LDH)in brain were determined by using the biochemical methods.The changes of brain water content in each group were measured by specific gravity method.The expression of aquaporin1(AQP1)and aquaporin4(AQP4)in brain tissues of rats in each group was observed by immunohistochemistry.【Results】Compared with the hypoxic model group,rosamultin could significantly increase the NO content and SOD activity in plasma(P<0.01),reduce MDA content in plasma(P<0.01)and LDH activity(P<0.01),water content(P<0.05)and the number of AQP1 and AQP4 positive cells in rat brain tissues with acute hypobaric hypoxia(P<0.01).【Conclusion】Rosamultin can improve the antioxidant ability and effectively reduce the degree of brain edema,so as to protect the brain injury induced by acute hypobaric hypoxia in rats.
