祛风止咳方对感染后咳嗽模型大鼠神经激肽-1受体表达的影响

摘要 目的:分析感染后咳嗽模型大鼠神经肽P物质(SP)、神经激肽-1受体(NK-1R)在祛风止咳方的作用下发生的改变,探究在感染后咳嗽方面,该方的作用机理。方法:将大鼠(40只)随机分成4组:西药治疗组,中药治疗组、模型组、空白对照组,构建PIC模型:雾化吸入辣椒素、脂多糖(LPS)滴鼻模拟感染革兰氏阴性菌,各组大鼠给予灌胃处理7d后,获取肺组织,研究其病理学变化,采用ELLSA法检测SP含量,免疫组化法、半定量评分研究肺组织中SP改变,肺组织中表达的NK-1R用Western-Blot进行检测。结果:(1)SP含量的分析:模型组较空白对照组具有更高的SP含量(P<0.05);无论是中药治疗组还是西药治疗组肺组织中较模型组具有更低的SP含量,下降显著的是中药治疗组(P<0.05);(2)SP免疫组化结果:中药治疗组和西药治疗组较模型组表达更低的SP蛋白,降低更明显的是中药治疗组(P<0.05);(3)NK-1R的Western-Blot检测结果:中药治疗组和西药治疗组肺组织较模型组表达更低的NK-1R蛋白,降低更显著的是中药治疗组。结论:祛风止咳方能够降低大鼠肺组织中SP及NK-1受体蛋白的表达,通过NK-1-SP途径来降低感染后咳嗽的气道神经源性炎症。 Objective:To analyze the changes of neuropeptide substance P(SP)and neurokinin-1 receptor(NK-1R)in cough model rats after infection under the action of Qufeng Zhike recipe,and to explore the mechanism of this prescription in post-infection cough.Methods:40 rats were randomly divided into 4 groups:Western medicine treatment group,traditional Chinese medicine treatment group,model group and blank control group.PIC model was established:aerosol inhalation of capsaicin and lipopolysaccharide(LPS)nasal drip simulated infection of Gram-negative bacteria.After intragastric administration for 7 days,lung tissue was obtained to study its pathological changes.SP content was detected by ELLSA method,immunohistochemistry and semi-quantitative score were used to study the changes of SP in lung tissue.The expression of NK-1R in lung tissue was detected by Western-Blot.Results:The analysis of SP content:the SP content in the model group was higher than that in the blank control group,and the SP content in the lung tissue of the traditional Chi-nese medicine and western medicine treatment group was lower than that of the model group,and the decrease was significant in the traditional Chinese medicine treatment group and western medicine treatment group(P<0.05).Immunohistochemical results of traditional Chinese medicine SP:the expression of SP protein in the traditional Chinese medicine treatment group was lower than that in the model group,and the decrease was more obvious in the traditional Chinese medicine treatment group(P<0.05).The results of Western-Blot detection of NK-1R:the expression of NK-1R protein in lung tissue of traditional Chinese medicine and western medicine treatment group was lower than that of model group,and the decrease was more significant in traditional Chinese medicine treatment group.Conclusion:Qufeng Zhike recipe can reduce the expression of SP and NK-1 receptor protein in lung tissue of rats,and reduce the airway neurogenic inflammation of cough after infection through NK-1-SP pathway.
