摘要
目的:通过网络药理学探讨二陈汤治疗非酒精性脂肪肝(NAFLD)的作用机制。方法:使用TCMSP预测二陈汤可能的有效成分和目标靶点,并通过DisGeNET、GeneCard数据库搜索和筛选NAFLD的相关靶点。使用STRING数据库和靶点共同预测目标蛋白相互作用关系,运用Cytoscape软件进行可视化并筛选出10个关键靶点,并基于DAVID数据库进行GO功能以及KEGG通路富集分析。最后将二陈汤与NAFLD的共同预测靶点用Cytoscape建立“成分-靶点-通路”作用网络。结果:(1)筛选出123个二陈汤的活性成分、251个药物预测靶点和NAFLD疾病相关靶点189个,二陈汤与NAFLD的共同靶点44个。(2)基于STRING数据库构建蛋白-蛋白相互作用网络,并筛选出IL6、TNF、JUN、AKT1、PRARG、MAPK8、IL10、CXCL8、CASP3、IL1B关键靶点。(3)在二陈汤作用于NAFLD的机制中,富集分析揭示了总共15个生物学过程、3个细胞学成分、5个分子功能和24条KEGG途径。其主要信号通路包括NAFLD、MAPK、FoxO、胰岛素抵抗、AMPK等,主要涉及RNA转录、细胞凋亡、DNA转录、炎症反应、基因表达、细胞内脂多糖反应、细胞增殖等多个生物学进程。结论:从网络药理学的角度来看,二陈汤可能在NAFLD的治疗中具有多种途径和多个靶点,为进一步揭示其作用机理提供参考,也为NAFLD的治疗提供了生物信息学基础。
Objective:To explore the mechanism of Erchen decoction in the treatment of nonalcoholic fatty liver disease(NAFLD)through network pharmacology.Methods:TCMSP was used to predict the possible active ingredients and target targets of Erchen Tang,and the relevant targets of NAFLD were searched and screened through DisGeNET and GeneCard databases.The STRING database and target were used to jointly predict the interaction relationship between target proteins.The CYTOSCAPE software was used to visualize and screen 10key targets,and the DAVID database was used to perform GO function and KEGG pathway enrichment analysis.Finally,the joint predic-tion target of Erchen Tang and NAFLD was used to establish the action network of"original component target original pathway"with Cytoscape.Results:(1)123 pieces of Erchen soup were screened Active ingredients,251 drug prediction targets,189 NAFLD disease related targets,and 44 common targets of Erchen Decoction and NAFLD.(2)Building Eggs Based on STRING Database Leukogen protein interaction network,and key targets of IL6,TNF,JUN,AKT1,PRARG,MAPK8,IL10,CXCL8,CASP3,and IL1B were screened.(3)The effect of Erchen Decoction on In the mechanism of NAFLD,enrichment analysis revealed a total of 15 biological processes,3 cytological com-ponents,5 molecular functions and 24 KEGG pathways.Its main signal paths include NAFLD,MAPK,FoxO,insulin resistance,AMPK,etc.,mainly involve RNA transcription,apoptosis,DNA transcription,inflammatory reaction,gene expression,intracellular lipopolysaccha-ride reaction Cell proliferation and other biological processes.Conclusion:From the perspective of network pharmacology,Erchen decoction may have multiple pathways and targets in the treatment of NAFLD,providing a reference for further revealing its mechanism of action,and also providing a bioinformatics basis for the treatment of NAFLD.
