肺功能检查在博来霉素诱导的大鼠肺纤维化中的实验研究 被引量:1

摘要 目的由于条件限制,在许多大鼠疾病模型中无法检测肺功能的变化情况,检测肺纤维化大鼠的肺功能对实验动物的肺功能应用具有重要意义。方法12只健康SD大鼠,随机分成正常对照组和博来霉素组,气管注射博来霉素(5 mg/kg)复制大鼠肺纤维化模型。于第28日腹主动脉放血法处死,比较两组体质量及肺质量,计算肺系数,检测两组大鼠肺功能和肺组织中羟脯氨酸的含量。结果与正常对照组比较,博来霉素组大鼠肺系数明显升高,博来霉素组羟脯氨酸的含量明显升高,同时HE和MASSON病理染色提示大鼠肺纤维化模型复制成功。大鼠肺功能检查结果:用力肺活量(forced vital capacity,FVC)、50%肺活量的呼气流速(forced expiratory flow,FEF)、最大呼气流量(peak expiratory flow,PEF)均较正常对照组明显降低,且具有统计学意义,另外,博来霉素组FEV0.1/FVC较正常对照组升高。结论大鼠肺纤维化模型中肺功能检测技术基本成熟,可以进一步扩大样本量进行研究。 Objective Because of the limitation of conditions;it is impossible to detect the changes of pulmonary function in many rat disease models.Detection of pulmonary function in rats with pulmonary fibrosis is of great significance to scientific research and clinical prevention and treatment of pulmonary fibrosis.Methods Healthy SD rats were randomly grouped according to random number table.Rats were induced pulmonary fibrosis by intratracheal injection of bleomycin.The rats with pulmonary fibrosis were randomly divided into two groups:control group,namely rats receiving intratracheal injection of saline;model group,namely rats receiving intratracheal injection of BLM;By analyzing the general conditions of the rats,lung coefficients were calculated,and lung functions of the rats in each group were examined.The H&E and Mason’s staining were performed to evaluate pulmonary fibrosis injury,inflammatory cell infiltration,structural disorder and collagen deposition.The contents of Hydroxyproline was determined by biochemical kits.Results BLM-treated rats exhibited typical symptoms of pulmonary fibrosis,featured as increased collagen deposition and alveolitis,and fibrosis and the parenchymal structural disorder in the lung.Compared with the bleomycin group,the lung coefficient,lung function and hydroxyproline in the hydrogen inhalation intervention group were significantly reduced,with statistical differences.The results of pulmonary function test in rats were as follows:FVC(forced vital capacity)、50%FEF(forced expiratory flow)、PEF(peak expiratory flow)were significantly lower than those in the normal control group.And it has statistical significance.Conclusions The pulmonary function detection technology in rat pulmonary fibrosis model is mature,which can further expand the sample size for research.
