摘要
本实验采用索氏提取辅助超声技术,以AB-8大孔吸附树脂为分离介质,研究提取连翘有效成分的最佳工艺条件。结果显示:提取剂浓度为80%,回流4次,回流温度92℃时,提取率最高(连翘苷3.988%,芦丁2.452%,连翘脂苷6.122%);改变提取剂的pH值可导致连翘苷转变为连翘次生苷;最佳上样流速为2BV/h,洗脱流速为3BV/h,连翘苷的最佳洗脱剂浓度为50%乙醇(纯度达95%),芦丁和连翘脂苷最佳洗脱剂浓度为30%乙醇(纯度达93%)。
In this experiment,the active ingredients of forsythia are extracted by soxhlet extraction assisted ultrasonic technology.Using AB-8 macroporous adsorption resin as separation medium,the results shown that extracting concentration 80%,the back four times and reflux temperature 92℃have the highest extraction yield(forsythia 3.988%,rutin 2.452%,forsythia A 6.122%);pH value leads to the conversion of forsythia into secondary glycoside;when active ingredients of forsythia is separated by AB-8 macroporouswith the optimal flow rate 2 BV/h,elution velocity 3 BV/h,the optimum concentration of forsythia suspense is 50%(purity up to 95%),the rutin and forsythia 30%(purity up to 93%).
