基于网络药理学数据挖掘探讨中药复方治疗前列腺癌的作用机制

摘要 目的基于网络药理学数据挖掘临床研究中运用的中药复方,探讨其治疗前列腺癌(prostate cancer,PCa)的作用机制。方法基于网络药理学研究PCa的临床治疗中运用的中药复方活性成分、药物作用靶点集作用通路。通过中国知网(China National Knowledge Infrastructure,CNKI)筛选治疗PCa复方;通过中医传承计算平台(Chinese herbal compound Inheritance computing platform,TCMiCS)统计分析药物;通过美国国家生物信息技术中心的高通量基因表达(Gene Expresion Omnibus,GEO)数据库筛选前列腺癌的疾病靶点;通过中药系统药理学数据库与分析平台(traditional Chinese medicine systems pharmacology database and analysis platform,TCMSP)数据筛选治疗PCa药物。进而对这些靶点进行基因本体论(gene ontology,GO)富集和京都基因和基因组编码(Kyoto Encyclopedia of Genes and Genomes,KEGG)通路富集分析,从而揭示现阶段中医药治疗PCa的作用机制。结果共纳入160味药517药次,其中出现8次及以上的共16味,其中黄芪、熟地黄、姜黄出现频次排名前三位。从152个活性成分中找到对应PCa病变的35个靶点。GO富集分析得到612个生物过程(biological process,BP)、53个细胞组成(cellular component,CC)、37个分子功能(molecular function,MF)富集通路,KEGG富集分析得到最显著富集的20个通路。结论网络药理学为阐明中药复发治疗PCa作用机制提供了新方法,有助于中医药现代化研究的开展。 Objective To explore the mechanism of the Chinese herbal compound used in clinical research based on the data of network pharmacology.Methods The active components of Chinese herbal compotind and the target set pathway of drug action were used in the clinical research of prostate cancer.The compoimd of prostate cancer was screened by China National Knowledge Infrastructure(CNKI).The Chinese herbal compound Inheritance computing platform(TCMiCS)was used to analyze the drugs.The target of prostate cancer was screened by Gene Expresion Omnibus(GEO)database of National Center for biotechnology.The treatment of prostate cancer was screened by traditional Chinese medicine systems pharmacology database and analysis platform(TCMSP)data.Then,the purpose of this study is to analyze the concentration of gene ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway to reveal the mechanism of the treatment of prostate cancer by Chinese herbal compound.Results A total of 160 herbs and 517 prescriptions were included,among which 16 herbs appeared 8 times or more.Among them,Huangqi,Shudihuang,and Jianghuang were the top 3 herbs with the highest frequency of appearance.From 152 active ingredientB,35 targets were identified for prostate cancer lesions.Gene ontology(GO)enrichment analysis revealed 612 enriched biological processes(BP),53 cellular components(CC),and 37 molecular functions(MF)pathways.KEGG enrichment analysis identified the top 20 significantly enriched pathways.Conclusion Network pharmacology provides a new approach to elucidate the mechanism of traditional Chinese medicine in the recurrence treatment of prostate cancer,which is helpful for the modernization of traditional Chinese medicine research.
