基于激痛点不同干预方法对MPS大鼠模型脊髓背角星型胶质细胞及神经生长因子影响 被引量:8

Effects of Different Intervention Methods on Astrocytes and Nerve Growth Factor in the Spinal Dorsal Horn of MPS Rats

摘要 目的观察针刺、艾灸两种不同干预方法对MPS大鼠模型脊髓背角星形胶质细胞、神经生长因子(NGF)的影响,探讨针刺、艾灸治疗MPS的中枢镇痛机制。方法将32只SD大鼠采用随机表法分为空白组、模型组、针刺组、艾灸组4组,造模方式采用顿性打击结合离心运动的方法,建立MPS大鼠模型。12周建模结束后,模型组不做治疗干预,针刺组、艾灸组进行相应的治疗。以热板实验法检测各阶段大鼠热缩足潜伏期,HE染色观察激痛点局部组织的病理变化,采用免疫组化检测法检测大鼠脊髓背角星型胶质细胞原纤维酸性蛋白GFAP免疫阳性细胞表达,Wes全自动蛋白质印迹定量检测脊髓背角神经生长因子NGF蛋白表达量。结果治疗后,同空白组比较,模型组、针刺组、艾灸组大鼠热缩足潜伏期降低(P<0.05),脊髓背角GFAP阳性细胞数目增多(P<0.05),NGF蛋白表达增加;同模型组比较,针刺组、艾灸组大鼠热缩足潜伏期增高(P<0.05),GFAP阳性细胞数目减少(P<0.05),NGF蛋白表达降低(P<0.05),且针刺组效果更佳。结论针刺、艾灸激痛点治疗MPS大鼠均可提高其热缩足潜伏期,减轻激痛点局部组织病理变化,降低脊髓背角星形胶质细胞原纤维酸性蛋白GFAP、神经生长因子的含量,推测其中枢镇痛机制可能与抑制脊髓背角星形胶质细胞的活化,降低NGF表达有关。 Objective To observe the effects of acupuncture and moxibustion on astrocytes and nerve growth factor(NGF)in spinal dorsal horn of MPS rats,and to explore the central analgesic mechanism of acupuncture and moxibustion on MPS.Methods 32 SD rats were randomly divided into four groups:blank group,model group,acupuncture group and moxibustion group.After 12 weeks of modeling,the model group received no treatment intervention,while the acupuncture group and moxibustion group received corresponding treatment.The hot plate test was used to detect the latent period of rat’s hot feet at each stage.HE staining was used to observe the pathological changes of local tissues at the pain point.The expression of GFAP immunoreactive cells in astrocytes of spinal dorsal horn was detected by immunohistochemistry.The expression of NGF protein in spinal dorsal horn was quantitatively detected by Wes automatic Western blot.Results After treatment,compared with the blank group,the latent period of heat contraction foot in model group,acupuncture group and moxibustion group decreased(P<0.05),the number of GFAP positive cells in spinal dorsal horn increased(P<0.05),and the expression of NGF protein increased;compared with the model group,the latent period of heat contraction foot in acupuncture group and moxibustion group increased(P<0.05),the number of GFAP positive cells decreased(P<0.05),and the expression of NGF protein increased and the effect of acupuncture group was better.Conclusion Acupuncture and moxibustion can improve the latent period of heat contraction foot,reduce the local histopathological changes of pain stimulation point,and reduce the contents of GFAP and NGF in astrocytes of spinal dorsal horn in MPS rats.It is speculated that the central analgesic mechanism may be related to inhibiting the activation of astrocytes and reducing the expression of NGF in spinal dorsal horn.
