基于网络药理学探讨补中益气汤治疗重症肌无力分子机制 被引量:6

Mechanism of Buzhong Yiqi Decoction(补中益气汤)for Treating Myasthenia Gravis Based on Network Pharmacology

摘要 目的运用网络药理学方法探讨补中益气汤治疗重症肌无力(myasthenia gravis,MG)的潜在作用机制。方法通过DisGeNET、TTD、Drugbank、GeneCards及OMIM数据库检索获取MG疾病的相关靶点,利用中药系统药理学数据库与分析平台(TCMSP)获得补中益气汤的化学成分和作用靶点,采用R软件对药物和疾病靶点进行匹配映射,使用String数据库构建靶点蛋白质互作网络(PPI),利用R软件对交集靶点进行GO生物过程分析和KEGG通路富集分析,并使用R软件及Cytoscape软件对结果进行绘图可视化。结果通过筛选得到补中益气汤133个主要活性成分,共涉及258个作用靶点,作用于MG疾病的靶标67个,可能通过AGE-RAGE信号通路、IL-17信号通路、TNF信号通路、T细胞受体信号通路、Th17细胞分化信号通路、Toll样受体等信号通路而发挥治疗MG的作用。结论补中益气汤对MG的治疗作用体现了中药多成分、多靶点、多途径的特点,为进一步阐释其治疗MG的作用机制与临床应用提供了科学依据。 Objective To investigate the potential molecular mechanism of Buzhong Yiqi Decoction(补中益气汤)for treating myasthenia gravis(MG)based on network pharmacology.Methods MG disease targets were collected from DisGeNET,TTD,Drugbank,GeneCards and OMIM databases,the chemical components of Buzhong Yiqi Decoction and their corresponding targets were obtained from the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP).Targets of drugs and disease were matched with R software.The protein-protein interaction(PPI)network was constructed by String.The Gene Ontology(GO)enrichment and the Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway were analyzed using by R software,and the results were visualized by Cytoscape software.Results Through screening,67 main active components of Buzhong Yiqi Decoction were obtained,involving a total of 258 targets,and 67 targets were related to the MG disease.They may played an therapeutic role in treating MG by AGE-RAGE pathway,IL-17 pathway,TNF pathway,T cell receptor pathway,Th17 cell differentiation pathway,Toll-like receptor pathway and soon.Conclusion The therapeutic effect of Buzhong Yiqi Decoction on MG reflects the multi-component,multi-target and multi-pathway characteristics of traditional Chinese medicine,which provides a scientific basis for further explaining its mechanism and clinical application on MG.
