摘要
目的基于网络药理学技术探讨乌头汤(WTT)治疗类风湿关节炎(RA)的作用机制。方法通过在TCMSP平台对乌头汤进行活性成分筛选,在DrugBank数据库进行靶点预测,应用Cytoscape 3.7.1软件构建成分-靶点网络;在OMIM、TTD及DisGenet数据库中进行检索,获取疾病靶点;构建韦恩图,取疾病与药物靶点交集,得到乌头汤治疗RA的作用靶点;运用ClueGO插件完成GO分析和KEGG分析。结果共得到乌头汤中144种有效成分及134个潜在靶点,获取RA疾病相关靶点1896个,乌头汤-RA疾病共同靶点69个,GO功能分析共包含354条富集结果,其中生物过程294条,分子功能52条,细胞组成8条,KEGG分析共筛选出57条通路在治疗RA方面具有作用。结论乌头汤可能是调节PTGS2、ESR1、ESR2、NOS2、AR等靶点,调控TNF信号通路、人巨细胞病毒感染、IL-17信号通路、Th17细胞分化、NOD样受体信号通路、Th1和Th2细胞分化、T细胞受体信号通路、FOXO信号通路、Toll样受体信号通路等通路,从而抑制炎症,调节免疫功能达到治疗RA的目的。
Objective To investigate the mechanism of Wutou Decoction(乌头汤,WTD)in the treatment of rheumatoid arthritis(RA)based on network pharmacology. Methods The active ingredient was screened on the TCMSP platform,the target was predicted in the DrugBank database,the component-target network was constructed by Cytoscape 3.7.1 software,and the target was obtained by searching in the OMIM,TTD and DisGenet databases. The Wayne map was constructed,and the intersection of disease and drug targets was obtained,and the target of treatment of RA with Wutou Decoction was obtained. GO analysis and KEGG analysis were performed using ClueGO plug-in. Results A total of 144 active constituents and 134 potential targets were obtained from Wutou Decoction. 1896 RA-related targets were obtained,69 common targets of WTD-RA disease,and GO functional analysis contained 354 enrichment results. Among them,there were 294 biological processes,52 molecular functions,and 8 cell components. A total of 57 pathways were screened by KEGG to play a role in the treatment of RA. Conclusion WTD may be the target of regulating PTGS2,ESR1,ESR2,NOS2,AR,etc. The pathway of action may be through TNF signaling pathway,human cytomegalovirus infection,IL-17 signaling pathway,Th17 cell differentiation,NOD-like receptor signaling pathway,Th1 and Th2 cell differentiation,T cell receptor signaling pathway,FOXO signaling pathway,Toll-like receptor signaling pathway and other pathways,thereby inhibiting inflammation and regulating immune function to achieve the purpose of treating RA.
