白芥子散治疗哮喘网络药理学作用机制研究 被引量:7

摘要 目的基于网络药理学的方法探讨白芥子散治疗哮喘的可能作用机制。方法应用中医药系统药理学数据库和分析平台(TCMSP)及Uniprot数据库筛选出白芥子散有效成分及靶点基因;同时从OMIM、DisGeNET和GeneCards数据库获取哮喘相关靶点;筛选得到疾病-药物成分共同靶蛋白,同时使用String数据及Cytoscape插件CytoNCA对该网络进行拓扑学分析得到核心靶点;对核心靶点基因利用Omicshare数据库进行GO分析和KEGG分析。结果从白芥子散中筛选出68个化学成分,涉及治疗哮喘的12个靶点;根据拓扑分析筛选出核心靶点29个;GO分析结果表明:生物过程主要涉及细胞因子介导的信号通路、炎性反应、防御反应、免疫反应等方面;分子功能主要涉及细胞因子活性、细胞因子受体结合、信号受体结合、生长因子受体结合等方面;细胞组成主要涉及细胞外空间、质膜外侧、细胞外区域等方面;KEGG分析结果表明主要涉及细胞因子-细胞因子受体相互作用、炎性反应性肠病(IBD)、IL-17信号通路、TNF信号通路等信号通路。结论该研究体现了白芥子散通过多靶点、多通路治疗哮喘,为以后科研以及临床研究提供理论依据。 Objective To explore the possible mechanism of Baijiezi Powder(白芥子散)in the treatment of asthma based on network pharmacology.Methods The active constituents and target genes of Brassica campestris were screened using the Chinese Medicine System Pharmacology Database and Analysis Platform(TCMSP)and Uniprot database.The asthma-related targets were obtained from the OMIM,DisGeNET and GeneCards databases.The disease-drug composition was screened.The common target protein,using String data and Cytoscape plug-in CytoNCA to perform topology analysis on the network to obtain the core target;the core target gene using Omicshare database for GO analysis and KEGG analysis.Results 68 chemical components were screened from Baijiezi Powder,involving 12 targets for asthma treatment;29 core targets were screened by topological analysis;GO analysis showed that biological processes mainly involved cytokine-mediated signaling pathways,inflammatory response,defense response,immune response,etc.;molecular function mainly involved cytokine activity,cytokine receptor binding,signal receptor binding,growth factor receptor binding,etc.;cell composition mainly involved extracellular space,outer side of plasma membrane,extracellular region,etc.;KEGG analysis results indicated that it mainly involves cytokine-cytokine receptor interaction,inflammatory bowel disease(IBD),IL-17 signaling pathway,TNF signaling pathway and other signaling pathways.Conclusion This study demonstrates that Baijiezi Powder can treat asthma through multi-target and multi-channel treatment,providing theoretical basis for future research and clinical research.
