补阳还五汤抗ApoE-/-小鼠动脉粥样硬化作用与机制研究 被引量:3

摘要 目的探究补五还阳汤对ApoE-/-小鼠动脉粥样硬化的抗击作用与机制研究。方法构建ApoE-/-动脉粥样硬化模型,并分为造模组、观察组A和观察组B各5只,分别每天给予高脂饮食、高脂饮食+补五还阳汤5 g/kg、补五还阳汤15 g/kg共12周,并用C57BL/6J-12小鼠5只作为空白对照进行正常饮食,对比4组小鼠的体质量、血脂、炎性因子的水平。结果给药前4组小鼠体质量并无明显差异(P<0.05),随着时间增加,各组小鼠体质量均明显增大,对比给药12周后各组小鼠的体质量,发现造模组和观察组小鼠体质量明显低于空白对照组(P<0.05),而观察组A小鼠体质量明显低于造模组小鼠,观察组A与观察组B小鼠体质量并无明显的差异(P>0.05);造模组小鼠的甘油三酯(TG)、总胆固醇(TC)、低密度蛋白胆固醇(LDL-C)、氧化低密度脂蛋白(ox-LDL)水平明显高于空白对照组小鼠(t=5.274、5.101、4.763、4.856,P<0.05),而高密度脂蛋白(HDL-C)、载脂蛋白(ApoA-I)显著低于空白对照组小鼠(t=5.924、5.806,P<0.05);观察组A小鼠的TG、TC、LDL-C、ox-LDL水平明显低于造模组小鼠(t=6.283、5.934、5.553,P<0.05),而HDL-C、ApoA-I显著高于造模组小鼠(t=4.683.5.271,P<0.05),而观察组A与观察组B小鼠并无明显差异(P>0.05);造模组小鼠血清中白细胞介素-1(IL-1)、白细胞介素-6(IL-6)、白细胞介素-10(IL-10)、肿瘤坏死因子α(TNF-α)、超敏C反应蛋白(hs-CRP)、细胞间黏附因子1(ICAM-1)、血管细胞黏附因子1(VCAM-1)、单核细胞趋化活性蛋白1(MCP-1)的含量均明显高于空白对照组小鼠(t=9.238、12.156、7.232、15.681、21.164、11.059、13.068、16.329,P<0.05),而观察组A小鼠以上指标均明显低于造模组小鼠(t=7.435、10.182、6.568、13.143、19.642、10.506、12.306、13.132,P<0.05),观察组A与观察组B并无明显差别(P>0.05);而4组小鼠NF-κB的表达水平并无明显变化。结论补五还阳汤可以降低ApoE-/-小鼠的体质量、血脂水平和炎性因子水平,但对NF-κB无影响。 Objective To explore the anti-allergic effect and mechanism of Buyang Huanwu Decoction on ApoE-/-mice atherosclerosis.Methods The ApoE-/-atherosclerosis model was constructed and divided into three groups:model group,observation group A and observation group B were each given a high-fat diet daily,high-fat diet+Buyang Huanwu Decoction 5 g/kg,high-fat diet+Buyang Huanwu Decoction 20 g/kg for 12 weeks,and 5 normal C57BL/6J-12 mice were selected as a blank control,then compare the body weight,blood lipids and inflammatory factors of the four groups of mice.Results There was no significant difference in body weight between the four groups before administration(P<0.05).The body weight of each group increased significantly with the increase of time.The body weight of each group was compared after 12 weeks of administration.The weight of the mice in the observation group was significantly lower than that in the blank control group(P<0.05),while the weight of the mice in the observation group A was significantly lower than that in the model group.There was no significant difference in the body weight between the observation group A and the observation group B mice(P>0.05);the levels of TG,TC,LDL-C and ox-LDL in the model mice were significantly higher than those in the blank control group(t=5.274,5.101,4.763,4.856,P<0.05),while HDL-C and ApoA-I were significantly lower than the blank control group(t=5.924,5.806,P<0.05);the levels of TG,TC,LDL-C and ox-LDL in the observation group A were significantly lower than those in the model group(t=6.283,5.934,5.553,P<0.05),while HDL-C and ApoA-I were significantly higher than model mice(t=4.683.5.271,P<0.05),while there was no significant difference in observation group A and observation group B mice(P>0.05);serum IL-1,IL-6,IL-10,TNFα,hs-CRP,ICAM-1,VCAM-1,MCP-1 in the serum of model mice.The content was significantly higher than that of the blank control group(t=9.238,12.156,7.232,15.681,21.164,11.059,13.068,16.329,P<0.05),while the above indicators in the observation group A mice were significantly lower than the model mice(t=7.435,10.182,6.568,13.143,19.642,10.506,12.306,13.132,P<0.05),there was no significant difference between observation group A and observation group B(P>0.05);while the expression levels of NF-κB in the four groups of mice did not change significantly.Conclusion Buyang Huanwu Decoction can reduce the body weight,blood lipid level and inflammatory factor levels of ApoE-/-mice,but has no effect on NF-κB.
