针药并举疗法对阿尔茨海默病小鼠模型神经病理影响 被引量:1

Effect of Acupuncture and Medicine Combined Therapy on Neuropathology in a Mouse model of Alzheimer’s Disease

摘要 目的基于"肾脑相济"理论,通过观察针药并举疗法对阿尔茨海默病小鼠模型(快速老化小鼠)SAMP8大脑皮层β-淀粉样蛋白(β-amyloid protein,Aβ)、磷酸化tau蛋白(P-tau)及日常行为执行能力的影响,探讨中医药对阿尔茨海默病(Alzheimer’s Disease,AD)的治疗作用。方法选用30只清洁级7月龄雄性SAM种系小鼠,其中24只快速老化SAMP8小鼠以及6只抗快速老化SAMR1小鼠,将24只SAMP8小鼠随机分为模型组、针灸组、中药组、针药组,每组6只。使用6只同月龄SAMR1小鼠作为对照组。针灸组针刺"百会""肾俞""三阴交"穴;中药组给予补肾活血汤灌胃;针药组给予补肾活血汤灌胃同时针刺"百会""肾俞""三阴交"穴。各组均连续治疗4周。采用筑巢实验、免疫组化法及ELISA法观察不同中医疗法对SAMP8小鼠日常行为执行能力和神经病理改变的影响。结果与对照组相比,模型组SAMP8小鼠筑巢实验评分降低,大脑皮层内Aβ、P-tau蛋白含量明显增加(P<0.05);与模型组相比,针灸组、中药组、针药组SAMP8小鼠筑巢实验评分升高,大脑皮层内Aβ、P-tau蛋白含量明显下降(P<0.05),针灸组、中药组、针药组之间SAMP8小鼠筑巢实验评分,大脑皮层内Aβ、P-tau蛋白含量无明显差异,无统计学意义(P>0.05)。结论针药并举疗法对SAMP8小鼠Aβ及P-tau蛋白有较好的调控作用,提高筑巢实验评分,明显减少大脑皮层Aβ、P-tau蛋白含量,从而达到治疗阿尔茨海默病的目的。 Objective Based on the theory of"kidney and brain economy",through observing the acupuncture and medicine combined therapy for Alzheimer’s disease mouse model(rapidly aging mice)SAMP8 cerebral cortexβ-amyloid protein(Aβ)The effects of phosphorylated tau protein(P-tau)and daily behavioral performance on the therapeutic effects of traditional Chinese medicine on Alzheimer’s Disease(AD).Methods Thirty male 7-month-old male SAM germs were selected,including 24 fast-aging SAMP8 mice and 6 fast-aging SAMR1 mice.24 SAMP8 mice were randomly divided into model group,acupuncture group and Chinese medicine group.Acupuncture group,6 in each group.Six SAMR1 mice of the same age were used as a control group.The acupuncture group acupuncture the"Baihui""Shenshu"and"Sanyinjiao"points;the Chinese medicine group was given Bushen Huoxue Decoction;the acupuncture group was given Bushen Huoxue Decoction and acupuncture"Baihui"and"Shenshu""Sanyinjiao"points.Each group was treated continuously for 4 weeks.Nesting experiments,immunohistochemistry and ELISA were used to observe the effects of different Chinese medicine methods on daily behavior and neuropathological changes in SAMP8 mice.Results Compared with the control group,the nesting test scores of the SAMP8 mice in the model group were decreased,and the contents of Aβand P-tau in the cerebral cortex were significantly increased(P<0.05).Compared with the model group,the acupuncture group,the Chinese medicine group,and the needle The nesting test scores of SAMP8 mice in the drug group were increased,and the contents of Aβand P-tau protein in the cerebral cortex were significantly decreased(P<0.05).The nesting experiment scores of SAMP8 mice in the acupuncture group,Chinese medicine group and acupuncture group,there was no significant difference in the content of Aβand P-tau in the cortex,which was not statistically significant(P>0.05).Conclusion The combination of acupuncture and medicine has a good regulation effect on Aβand P-tau protein in SAMP8 mice,improve the nesting test score,and significantly reduce the content of Aβand P-tau in the cerebral cortex,so as to achieve the purpose of treating Alzheimer’s disease.
