摘要
目的观察电针心经对急性心肌缺血(AMI)大鼠海马DG区去甲肾上腺素(NE)、多巴胺(DA)及5-羟色胺(5-HT)的影响,探究海马DG区参与电针心经改善急性心肌缺血的作用机制。方法将雄性SD大鼠随机分成伪手术组、模型组、电针心经组(针刺组),每组6只。通过结扎冠状动脉左前降支,复制大鼠心肌缺血模型。电针心经组采用电针刺激"神门"-"通里"段,频率2 Hz,强度1 mA,刺激时间30 min,连续3 d。伪手术组、模型组不予电针。使用Powerlab16导生理记录仪记录心电图(ECG);用Masson染色法观察各组大鼠心肌纤维、胶原纤维变化;使用酶联免疫法(ELISA)检测血清肌酸激酶同工酶(CK-MB)含量;大鼠海马DG区NE、5-HT和DA的含量亦采用酶联免疫法检测。结果伪手术组存在大量心肌纤维,有少许胶原纤维;模型组出现大量胶原纤维呈网格状,也有一些分布松散的点状;针刺组显示胶原纤维分布范围减小。与伪手术组比较,模型组大鼠海马DG区5-HT、DA均明显降低,NE明显升高(P<0.05);与模型组比较,针刺组海马DG区NE显著降低,5-HT、DA均明显升高,且差异有统计学意义(P<0.05)。结论电针心经"神门"-"通里"经脉段对急性心肌缺血大鼠具有明显的协调保护效应,其作用机制与海马DG区单胺类神经递质的平衡有关。
Objective:To observe the effects of electroacupuncture on the treatment of norepinephrine(NE),dopamine(DA)and serotonin(5-HT)in the dentate gyrus of hippocampus in rats with acute myocardial ischemia(AMI).The mechanism of acute myocardial ischemia.Methods:Male Sprague-Dawley rats were randomly divided into a pseudo-surgery group,a model group,and an electroacupuncture group,with 6 rats in each group.The rat myocardial ischemia model was replicated by ligation of the left anterior descending coronary artery.The electroacupuncture group used electroacupuncture to stimulate the"Shenmen"-"Tongli"segment with a frequency of 2 Hz,an intensity of 1 mA,and a stimulation time of 30 min for 3 consecutive days.The sham operation group and the model group do not receive the electroacupuncture.Electrocardiogram(ECG)was recorded using a Powerlab 16 guided physiological recorder;The changes of myocardial fibers and collagen fibers were observed by Masson staining.The contents of NE,5-HT and DA in rat hippocampal DG were detected by enzyme-linked immunosorbent assay.Results:There are a large number of myocardial fibers in the sham operation group,and there is a little collagen fiber;In the model group,a large number of collagen fibers appeared in a grid shape,and some of them were loosely distributed.Acupuncture group shows a reduction in the distribution of collagen fibers.Compared with the sham operation group,the 5-HT and DA in the hippocampal DG area of the model group were significantly decreased,and the NE was significantly increased(P<0.05).Compared with the model group,the NE in the hippocampus DG area of the acupuncture group was significantly decreased,and 5-HT and DA were significantly increased,and the difference was statistically significant(P<0.05).Conclusion:Electroacupuncture through the"Shenmen"-"Tongli"meridian segment has obvious protective effect on acute myocardial ischemia rats,and its mechanism is related to the homeostasis of monoamine neurotransmitters in hippocampal DG.
