摘要
目的:采用3种大孔树脂对地龙酶解液进行纯化,筛选出最佳纯化树脂,并建立最佳纯化方法。方法:采用双酶酶解方法对地龙进行酶解得到地龙酶解液,同时对DA201-C、AB-8、D1013种大孔树脂进行考察,得到最佳吸附树脂,同时对树脂进行系统考察得到最佳吸附条件。结果:通过考察得出3种树脂的吸附率顺序为DA201-C>AB-8>D101,说明DA201-C树脂对地龙酶解液活性肽吸附效果最佳,同时对DA201-C树脂进行静态和动态吸附考察,得出最佳吸附条件为:酶解液浓度为20 mg/mL,调节pH至6.0,以2 BV/h的流速往DA201-C树脂上上样,以A220nm>0.10为上样终点,静态吸附3 h,最终有效成分吸附率为89.65%。结论:采用DA201-C树脂对地龙酶解液进行处理方法科学合理,利于后期实验进行。
Objective:To purify the enzymatic hydrolysate of earthworm with three macroporous resins and selecte the best purification method,obtained the optimum adsorption conditions.Methods:The enzymatic hydrolysate of earthworm was obtained by double enzymatic hydrolysis,and three kinds of macroporous resins,DA201-C,AB-8 and D101,were investigated to obtain the best adsorption resin.At the same time,the best adsorption conditions were obtained by systematic investigation of the resin.Results:The order of adsorption rate of the three resins was DA201-C>AB-8>D101,which indicated that DA201-C resin had the best adsorption effect on active peptide of earthworm enzymatic hydrolysate.Static and dynamic adsorption tests were carried out on DA201-C resin.The optimum adsorption conditions were as follows:concentration of enzymatic hydrolysate was 20 mg/mL,pH was adjusted to 6.0,flow rate of 2 BV/h was adjusted to sample DA201-C resin,and A220 nm was more than 0.1.The final effective component adsorption rate was 89.65%after static adsorption for 3 hours.Conclusion:DA201-C resin is a scientific and reasonable method for the treatment of enzymatic hydrolysate of earthworm,which is beneficial to the later experiment.
