复明颗粒对脑缺血再灌注模型大鼠脑额叶皮层细胞的影响

Effect of Fuming Granule on Brain Frontal Cortex Cells in Rats with Cerebral Ischemia-reperfusion Injury

摘要 目的:研究复明颗粒对大鼠脑缺血再灌注损伤的防治作用及作用机制。方法:85只SPF级Wistar大鼠分为假手术组、模型组、复明颗粒低剂量组(3.6 g生药/kg)、复明颗粒中剂量组(7.2 g生药/kg)、复明颗粒高剂量组(14.4 g生药/kg)、银杏叶片组(5.18 mg/kg)。预防灌胃给药7 d后,采用改良线栓法阻断大鼠左侧大脑中动脉供血区域局灶性脑缺血120 min后,恢复脑血流再灌注,继续灌胃给药治疗3 d。末次给药1 h后,观察复明颗粒对大脑缺血再灌注神经行为损伤评分、脑额叶皮层细胞尼氏染色、大鼠脑额叶Bax和Bcl-xl表达的影响。结果:复明颗粒高剂量组比模型组神经行为损伤评分显著降低,复明颗粒高剂量组额叶皮层细胞Bax表达比模型组显著降低,复明颗粒高剂量组Bcl-xl表达比模型组显著增加。结论:复明颗粒可通过调节神经元Bax和Bcl-xl表达对脑缺血再灌注模型产生保护作用。 Objective:To study the preventive and therapeutic effects of Fuming Granule on cerebral ischemia-reperfusion injury in rats.Methods:Eighty-five SPF-level Wistar rats were divided into sham operation group,model group,low-dose group of Fuming granules(3.6 g crude drug/kg),middle-dose group of Fuming granules(7.2 g crude drug/kg),and high-dose group of Fuming granules(14.4 g crude drug/kg),ginkgo leaf group(5.18 mg/kg).Seven days after the intragastric administration,the cerebral blood flow reperfusion was resumed by embolization of the left cerebral artery in the left cerebral artery for 120 minutes.The cerebral blood flow was reperfused and the drug was administered for 3 days.One hour after the last administration,the effects of Fuming Granule on the neurological behavioral damage of cerebral ischemia-reperfusion,Nissl staining of the frontal cortex,and the expression of Bax and Bcl-xl in the frontal lobe of rats were observed.Results:The high-dose group of Fuming granules was significantly lower than the model group.The expression of Bax in the frontal cortex cells of Fuming granules was significantly lower than that of the model group.The expression of Bcl-xl in the high-dose group of Fuming granules was significantly higher than that of the model group.Conclusion:Fuming Granule can protect the cerebral ischemia-reperfusion model by regulating the expression of Bax and Bcl-xl.
