活血潜阳祛痰方改善肥胖高血压大鼠心肌肥厚及对线粒体功能的保护作用

Protective Effect of Huoxue Qianyang Qutan Recipe(活血潜阳祛痰方)on Myocardial Hypertrophy and Mitochondrial Function in Obese Hypertensive Rats

摘要 目的观察活血潜阳祛痰方对肥胖高血压大鼠心肌肥厚及心肌线粒体功能的影响。方法5周龄雄性SPF级自发性高血压大鼠(SHR)适应性喂养1周后随机分为普通饲料组(SHR组,12只)、高脂饲料组108只,后者制备肥胖高血压大鼠模型,周龄/性别相匹配的SPF级正常血压京都种(WKY)大鼠喂养普通饲料(WKY组,12只)用作对照。10周后,造模成功的肥胖高血压大鼠随机分为模型组、西药(缬沙坦)组和中药(活血潜阳祛痰方)组。用药干预10周后行心超检测,测定心肌组织肥大基因脑钠肽(BNP)表达水平、线粒体氧化应激指标、线粒体电子传递链亚基蛋白表达水平和SIRT1/PGC-1α通路信号分子基因表达水平。结果与模型组比较:西药组和中药组大鼠左室收缩末期内径(LVIDs)明显增加(P<0.01),左心室后壁收缩期厚度(LVPWs)、室间隔收缩末期厚度(IVSs)明显降低(P<0.01);西药组和中药组BNP mRNA表达水平明显下调(P<0.05);西药组和中药组丙二醛(MDA)的含量明显下降(P<0.05),中药组谷胱甘肽过氧化物酶(GSH-Px)活性明显增加(P<0.05);西药组和中药组NDUFA13、SDHB蛋白表达水平明显上调(P<0.01);中药组心肌组织SIRT1、PGC-1α基因表达水平明显上调(P<0.05或P<0.01),西药组和中药组NRF1、TFAM基因表达水平明显上调(P<0.05)。结论活血潜阳祛痰方具有改善肥胖高血压大鼠心肌肥厚,保护心肌线粒体功能的作用。 Objective To observe the effect of Huoxue Qianyang Qutan Recipe(活血潜阳祛痰方,HQQR)on myocardial hypertrophy and myocardial mitochondrial function in obese hypertensive rats.Methods After 1 week of adaptive feeding,5-week-old male spontaneously hypertensive rats(SHR)were randomly divided into normal diet group(SHR group,n=12)and high-fat diet group(n=108).Obese hypertensive rats were established by the latter,and normal blood pressure Wistar-Kyoto(WKY)rats matched with age/sex fed with normal diet(WKY group,n=12)were as the control group.Ten weeks later,the obese hypertensive rats were randomly divided into model group,western medicine(valsartan)group and traditional Chinese medicine(HQQR)group.After drug intervention for 10 weeks,the echocardiography was performed and the expression levels of myocardial hypertrophy gene brain natriuretic peptide(BNP),mitochondrial oxidative stress indexes,mitochondrial electron transport chain subunit protein expressions and SIRT1/PGC-1αpathway signal molecule gene expressions were detected.Results Compared with those of the model group,the left ventricular end systolic diameter(LVIDs)in valsartan and HQQR groups increased significantly(P<0.01)while systolic thickness of left ventricular posterior wall(LVPWs)and end systolic septal thickness(IVSs)decreased significantly(P<0.01);the expression level of BNP mRNA in valsartan and HQQR groups decreased significantly(P<0.05);the content of malondialdehyde(MDA)in valsartan and HQQR groups decreased significantly(P<0.05)while the activity of glutathione peroxidase(GSH-Px)increased significantly in HQQR group(P<0.05).The expression levels of NDUFA13 and SDHB protein in valsartan and HQQR groups were both significantly up-regulated(P<0.01);the expression levels of SIRT1 and PGC-1αmRNA in myocardial tissues were significantly up-regulated in HQQR group(P<0.05 or P<0.01)and the expression levels of NRF1 and TFAM mRNA in valsartan and HQQR groups were significantly up-regulated(P<0.05).Conclusion HQQR could improve myocardial hypertrophy and protect myocardial mitochondrial function in obese hypertensive rats.
