当归芍药散对PM2.5致雄性大鼠生殖损伤的保护作用及其抗氧化机制 被引量:1

Protective and Antioxidant Effects of Danggui Shaoyao Powder(当归芍药散)in PM2.5-induced Reproductive Injury in Male Rats

摘要 目的评价PM2.5暴露的雄性生殖损伤及当归芍药散保护作用。方法30只雄性SD大鼠随机分为对照组、PM2.5组和当归芍药散低、中、高剂量组。第1~12周,各组大鼠(对照组除外)进行实时浓缩PM2.5暴露;第9-12周,对照组、PM2.5组予以生理盐水灌胃(2 mL/只),其余组给予相应剂量当归芍药散灌胃;第12周结束,观察大鼠肺系数、肺功能、睾丸系数、肺和睾丸病理及精子质量,检测血清生殖激素和睾丸氧化应激水平。结果PM2.5组用力肺活量(FVC)、0.3 s用力呼气容积(FEV0.3)较对照组降低、肺组织损伤评分升高(P<0.05,P<0.01);睾丸及附睾系数升高、睾丸组织Johnsen评分下降、附睾精子减少、畸形、活力显著降低(P<0.05,P<0.01);卵泡刺激素(FSH)、黄体生成素(LH)、泌乳素(PRL)、雌二醇(E)、睾酮(T)水平均降低(P<0.05,P<0.01);超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)显著降低,丙二醛(MDA)显著升高(P<0.05,P<0.01)。3种剂量当归芍药散均可不同程度减轻上述损伤,综合疗效以低剂量组较为显著。结论PM2.5暴露可导致雄性生殖激素紊乱、睾丸及附睾损伤,精子活力降低,氧化应激可能是其潜在机制。当归芍药散可通过纠正氧化/抗氧化失衡来改善PM2.5引起的雄性生殖损伤。 Objective To evaluate the effect of Danggui Shaoyao Powder(当归芍药散,DSP)on oxidative stress in male rats reproductive injury induced by particulate matter(PM)2.5.Methods Thirty male rats were randomly divided into control,model,low-DSP,medium-DSP and high-dose DSP groups.A real-time atmospheric concentrated PM2.5 exposure system was used to establish PM2.5-induced reproductive injury rats model.2 mL of 0.9%physiological saline were given by gavage in control and model groups from week 9 to week 12.Rats in treatment groups received DSP at relevant dosages.By the end of the 12 th week,lung coefficient,lung function and testis coefficient were detected.The lung tissue pathology,testicular tissue pathology and sperm morphology were observed and lung tissue injury score,Johnsen score and sperm quality analysis were performed.The levels of reproductive hormones was determined by ELISA,and the level of oxidative stress was measured using colorimetric method.Results Compared with those of the control group,the lung coefficient in PM2.5 group was increased(P<0.01).Lung tissue showed alveolar wall rupture,inflammatory cell infiltration,and the lung injury score was increased.The values of forced vital capacity(FVC)and 0.3 s forced expiratory volume(FEV0.3)were decreased(P<0.05,P<0.01).Testicular coefficient and epididymal coefficient increased.The level of spermatogenic cells was decreased,the arrangement was disordered,the number of sperm and sperm cells was decreased,and Johnsen score was decreased.The number of sperm in epididymal cavity was decreased.Sperm tail curl,sperm concentration and vitality was decreased significantly(P<0.01).The levels of follicle stimulating hormone(FSH),luteinizing hormon(LH),prolactin(PRL),estradiol(E2)and testosterone(T)were decreased(P<0.05,P<0.01).The levels of superoxide dismutase(SOD)and glutathione peroxidase(GSH-Px)were decreased(P<0.05,P<0.01),while the levels of malondialdehyde(MDA)were increased(P<0.01).DSP can alleviate above-mentioned injuries.The comprehensive evaluation of R value showed that the comprehensive curative effect of low dose DSP was more significant.Conclusion PM2.5 exposures can induce male reproductive hormone disorders,testis and epididymis injuries,and reduced sperm motility and concentration.The oxidative stress might be the underlying mechanism.DSP can improve PM2.5-induced male reproductive damages via correcting oxidant/antioxidant imbalance.
