基于HPLC测定不同产地西洋参中12种人参皂苷含量及指纹图谱研究 被引量:1

Determination of 12 Kinds of Ginsenosides in Xiyangshen(Panacis Quinquefolii Radix) from Different Origins and Fingerprints Based on HPLC

摘要 目的建立西洋参药材的指纹图谱,测定其中12种人参皂苷的含量,并进行化学模式识别分析。方法色谱柱:COSMOSIL 5C_(18)-MS-Ⅱ柱(4.6 mm×250 mm),流动相:乙腈(A)-0.1%磷酸水(B),流速:1.0 mL·min^(-1),检测波长:203 nm,柱温:40℃,进样量:10μL;利用“中药色谱指纹图谱相似度评价系统”(2004 A版)对西洋参样品色谱图进行相似度评价;利用SPSS及SIMCA软件对其进行聚类分析(HCA)、主成分分析(PCA)、偏最小二乘判别分析(PLS-DA)。结果28种不同产地西洋参药材的指纹图谱共检出12个共有峰,样品可分为4类,前4个主成分的贡献率达到84.849%;同时测定了28批西洋参药材中的12种(Rg1、Re、Rf、Rb1、Rc、Rg2、Ro、Rb2、Rb3、Rd、F2、Rg3)人参皂苷成分的含量,12种成分含量范围依次为:0.031~0.236 mg·g^(-1),0.737~1.769 mg·g^(-1),0.000~0.004 mg·g^(-1),1.036~3.396 mg·g^(-1),0.012~0.048 mg·g^(-1),0.113~0.324 mg·g^(-1),0.146~0.636 mg·g^(-1),0.065~0.162 mg·g^(-1),0.060~0.148 mg·g^(-1),0.170~0.627 mg·g^(-1),0.006~0.082 mg·g^(-1),0.000~0.292 mg·g^(-1)。结论可以利用HPLC指纹图谱技术结合统计学分析方法评价不同产地西洋参药材,更好的提高西洋参的临床应用价值。 Objective To establish the fingerprint of Xiyangshen(Panacis Quinquefolii Radix)medicinal materials,determine the contents of 12 kinds of ginsenosides and conduct chemical pattern recognition analysis.Method:COSMOSIL 5 C_(18)-MS-Ⅱcolumn(4.6 mm×250 mm)was used as the chromatographic column,acetonitrile(A)-0.1%phosphoric acid water(B)as the mobile phase,and the flow rate was 1.0 mL·min^(-1).The detection wavelength was 203 nm,the column temperature was 40℃,and the injection volume was 10μL.The similarity evaluation of the chromatograms of Xiyangshen(Panacis Quinquefolii Radix)samples was carried out using the Chinese Medicine Chromatographic Fingerprint Similarity Evaluation System(2004 A version).Use SPSS and SIMCA software were used to perform cluster analysis(HCA),principal component analysis(PCA)and partial least square discriminant analysis(PLS-DA).Results A total of 12 common peaks were detected in the fingerprints of 28 kinds of Xiyangshen(Panacis Quinquefolii Radix)medicinal materials from different origins.The samples can be divided into 4 categories,with 4 main components,and the contribution rate reached 84.849%.At the same time,12 kinds of 28 batches of Xiyangshen(Panacis Quinquefolii Radix)medicinal materials(Rg1,Re,Rf,Rb1,Rc,Rg2,Ro,Rb2,Rb3,Rd,F2,Rg3)were determined and the content ranges of 12 components were 0.031-0.236 mg·g^(-1),0.737-1.769 mg·g^(-1),0.000-0.004 mg·g^(-1),1.036-3.396 mg·g^(-1),0.012-0.048 mg·g^(-1),0.113-0.324 mg·g^(-1),0.146-0.636 mg·g^(-1),0.065-0.162 mg·g^(-1),0.060-0.148 mg·g^(-1),0.170-0.627 mg·g^(-1),0.006-0.082 mg·g^(-1)and 0.000-0.292 mg·g^(-1).Conclusion The established method for determining the contents of Xiyangshen(Panacis Quinquefolii Radix)is accurate,sensitive and reproducible.HPLC fingerprinting technology combined with statistical analysis methods can be used to evaluate Xiyangshen(Panacis Quinquefolii Radix)from different origins to better improve the clinical application value of Xiyangshen(Panacis Quinquefolii Radix).
