止痛消炎膏对慢性湿疹模型小鼠皮损及TEWL、SCH、湿度的影响

Effect of Zhitong Xiaoyan Ointment(止痛消炎膏)on Skin lesions,TEWL,SCH and Humidity of Chronic Atopic Dermatitis Model Mice

目的探究止痛消炎膏对慢性湿疹模型小鼠皮损及经表皮水分丢失(TEWL)、角质层含水量(SCH)、湿度的影响。方法40只Balb/c小鼠随机分为空白组、模型组、实验组和对照组,每组10只,采用2,4-二硝基氯苯(DNCB)诱导慢性湿疹模型,给药后通过EASI评分、HE染色观察小鼠皮肤外观及组织病理变化;利用GPSkin Barrier检测仪,检测干预0周、1周、2周TEWL、SCH、湿度等相关皮肤屏障功能指标的变化。结果干预2周后,实验组、对照组较模型组皮损症状减轻,镜下表皮增生、水肿、炎性细胞浸润情况有所缓解,且实验组的改善明显优于对照组;EASI评分及相关皮肤屏障功能指标结果显示:实验组和对照组的EASI评分、TEWL均显著低于模型组(P<0.05),SCH、皮肤湿度均显著高于模型组(P<0.05),实验组各项指标的改善均优于对照组(P<0.05)。结论止痛消炎膏对慢性湿疹小鼠模型具有一定的治疗作用,不仅能改善皮损,降低EASI评分,还能有效减少TEWL,增加SCH和湿度,修复皮肤屏障功能。

Objective To investigate the effects of Zhitong Xiaoyan Ointment(止痛消炎膏)on the skin lesions,transepidermal water loss(TEWL),stratum corneum hydration(SCH)and humidity of chronic atopic dermatitis model mice.Methods Forty Balb/c mice were randomly divided into the blank group,the model group,the experimental group and the control group,with 10 mice in each group.The chronic eczema model was induced by 2,4-dinitrochlorobenzene(DNCB).The skin appearance and histopathological changes of mice were observed by eczema area and severity index(EASI)score and HE staining after administration.GPSkin Barrier detector was used to detect the changes of TEWL,SCH,humidity and other related skin barrier function indexes after 0,1 and 2 weeks of intervention.Results After 2 weeks of intervention,compared with those of the model group,the symptoms of skin lesions in the experimental group and the control group were relieved,the conditions of epidermal hyperplasia,edema and inflammatory cell infiltration under the microscope were relieved and the improvement in the experimental group was significantly better than that in the control group.The results of EASI score and related skin barrier function indexes showed that the EASI score and TEWL of the experimental group and the control group were significantly lower than that of the model group(P<0.05).The SCH and skin humidity were significantly higher than those in the model group(P<0.05)and the improvement of each index in the experimental group was better than that in the control group(P<0.05).Conclusion Zhitong Xiaoyan Ointment has a certain therapeutic effect on chronic atopic dermatitis mice model,which can not only relieve the skin lesions,reduce the EASI score,but also effectively reduce TEWL,increase SCH and humidity and repair the skin barrier function.
