振腹环揉法对失眠大鼠下丘脑内SP、GAL、β-EP含量水平的影响 被引量:3

Effects of Abdominal Vibration and Ring-Kneading Manipulation on SP,GAL and β-EP in Hypothalamus of Insomnia Rats

摘要 目的通过检测失眠大鼠下丘脑内的SP、GAL、β-EP的浓度,结合失眠大鼠症状改善变化,探讨振腹环揉法治疗失眠的作用机制。方法选取40只SD大鼠,随机分为空白组、模型组、推拿组,每组10只大鼠。空白组不进行造模和治疗;模型组仅造模但不治疗;推拿组造模后推拿治疗。分别在治疗5 d、10 d各处死5只大鼠采用ELISA法检测失眠大鼠下丘脑内SP、GAL、β-EP浓度,并观察失眠大鼠症状改善情况。结果造模后模型组潜睡眠时间和睡眠持续时间均有显著性差异(P<0.01)。比较SP含量,第5天与第10天比较模型组、推拿组SP含量均升高(P<0.05)。比较治疗5 d、10 d后SP含量模型组小于其他各组,且存在统计学意义(P<0.05)。比较GAL含量,模型组、推拿组第5天和第10天比较,GAL含量均升高(P<0.05)。治疗5 d后和10 d后模型组GAL含量均低于其他各组,且存在统计学意义(P<0.05)。比较β-EP含量,第5天与第10天比较,模型组、推拿组β-EP含量均增高,有统计学意义(P<0.05)。比较治疗5 d后和10 d后β-EP含量各组间比较,模型组显著低于其他各组,有统计学意义(P<0.01)。结论PCPA造模法能够模拟大鼠失眠的发生,振腹环揉法具有改善失眠大鼠模型失眠症状的作用,振腹环揉推拿疗法能够调节失眠大鼠模型下丘脑内SP、GAL、β-EP浓度治疗失眠。振腹环揉推拿疗法能改善下丘脑AS细胞排列状态调节下丘脑功能,改善昼夜节律和下丘脑能量代谢。 Objective By detecting the levels of SP,GAL andβ-EP in the hypothalamus of insomnia rats,combined with the improvement of symptoms in insomnia rats,to explore the mechanism of vibration of the abdomen and ring-kneading manipulation in the treatment of insomnia.Methods Forty SD rats were selected and randomly divided into blank group,model group and Tuina group,with 10 rats in each group.The blank group did not undergo modeling or treatment.The model group was only modeled but not treated.The Tuina group was treated with Tuina after modeling.Five rats died in 5 and 10 days of treatment respectively.The levels of SP,GAL andβ-EP in the hypothalamus of insomnia rats were detected by ELISA method,and the improvement of symptoms of insomnia rats was observed.Results After modeling,there were significant differences in sleep duration and sleep duration in the model group(P<0.01).The SP level of the model group and the Tuina group increased on the 5 th and 10 th day(P<0.05).The SP level of the model group was smaller than that of the other groups after 5 and 10 days of treatment,and there was a statistical difference(P<0.05).The GAL level was increased in the model group and the Tuina group on the 5 th and the 10 th day(P<0.05).After 5 days of treatment and 10 days after treatment,the GAL level of the model group was lower than that of the other groups,and there was a statistical difference(P<0.05).The level ofβ-EP in the model group and the Tuina group was increased on the 5 th and 10 th day,and there was a statistical difference(P<0.05).Theβ-EP level between the models after 5 days of treatment and 10 days after treatment in the model group was significantly lower than that of the other groups with statistical differences(P<0.01).Conclusion PCPA modeling method can simulate the occurrence of insomnia in rats.Vibrating abdomen and ring kneading-method can improve the insomnia symptoms of insomnia rat models.Vibrating abdomen and ring-kneading therapy can regulate SP,GAL andβ-EP in the hypothalamus of insomnia rat models.Vibrating abdomen and ring-kneading therapy can improve the arrangement of hypothalamic AS cells,regulate hypothalamic function and improve circadian rhythm and hypothalamic energy metabolism.
