益气健脾抗癌法对结肠癌HT-29细胞自噬的作用研究 被引量:2

Effect of Invigorating Qi and Invigorating Spleen Anti-cancer Method on Autophagy of Colon Cancer HT-29 Cells

摘要 目的探索益气健脾抗癌法对体外培养的结肠癌HT-29细胞自噬作用及相关机制。方法将体外培养的结肠癌HT-29细胞分为高剂量益气健脾抗癌组,中剂量益气健脾抗癌组,低剂量益气健脾抗癌组,及不含药血清对照组。应用Western blot检测各组细胞中Beclin-1、LC3和p62蛋白表达量,吖啶橙染色检测细胞自噬。结果在作用48 h时,与不含药血清对照组相比,益气健脾抗癌法可激活HT-29细胞的自噬(P<0.05)。高剂量组较中、低剂量组对细胞的激活能力差异具有统计学意义,3个剂量组均可降低HT-29细胞的侵袭能力,高剂量组与其他组相比,差异有统计学意义(P<0.05)。结论益气健脾抗癌法可以激活HT-29细胞的自噬,且与剂量呈正相关。 Objective To explore the autophagy of colon cancer HT-29 cells cultured in vitro by invigorating Qi and strengthening spleen and anti-cancer method.Methods Colon cancer HT-29 cells cultured in vitro were divided into three groups:high-dose group,medium-dose group,low-dose group,and control group without drug serum.Western blot was used to detect beclin-1,LC3 and P62 protein expressions in each group,and acridine orange staining was used to detect autophagy.Results At 48 h of treatment,compared with the drug-free serum control group,the invigorating Qi and strengthening spleen and anti-cancer method could activate the autophagy of HT-29 cells(P<0.05).The activation ability of hT-29 cells in the high-dose group was statistically different from that in the middle-dose and low-dose groups.The invasion ability of HT-29 cells in the three dose groups was reduced,and the difference between the high-dose group and other groups was significant(P<0.05).Conclusion The invigorating Qi and strengthening spleen and anti-cancer method can activate the autophagy of HT-29 cells,which is positively correlated with the dose.
