针刺对帕金森抑郁大鼠血清胃饥饿素含量的影响

Effects of Mind-Regulating Acupuncture on Serum Ghrelin in Rats of Parkinson′s Disease with Depression

目的观察针刺对帕金森抑郁(PDD)模型大鼠血清中胃饥饿素(Ghrelin)含量的影响,探讨针刺治疗帕金森抑郁的作用机制。方法SD大鼠随机分为正常组、模型组、假手术组、西药组、针刺组,每组10只。采用单侧纹状体双靶点注射法和慢性不可预见性温和性应激(CUMS)法制备PDD模型。针刺组针刺"百会透太阳""中脘",每日治疗20 min;西药组予氟西汀6 mg/kg,多巴丝肼片6 mg/kg灌胃。28 d后进行蔗糖溶液偏好实验、旷场实验,采用酶联免疫吸附法(ELISA)检测血清Ghrelin的含量。结果与正常组比较,模型组大鼠的Ghrelin含量明显减少(P<0.01),西药组与模型组无明显差异,针刺组的Ghrelin含量均明显升高(P<0.05)。结论针刺对PDD模型大鼠的抑郁样行为均有明显的改善作用,并能提高其血清中Ghrelin的含量,这可能是针刺治疗PDD的作用机制之一。

Objective To observe the effect of acupuncture on the content of Ghrelin in serum of rats of Parkinson′s disease with depression(PDD)and explore the mechanism of acupuncture in the treatment of Parkinson′s disease with depression.Methods Fifty SD rats were randomly divided into the normal group,the model group,the sham operation group,the western medicine group,the diversions acupuncture group and the ordinary acupuncture group(n=10).The PDD model was established by unilateral striatal double target injection and chronic unpredictable mild stress(CUMS).The acupuncture was applied to Baihui(GV20),penetrating Taiyang(EX-HN5)and Zhongwan(RN12).Fluoxetine(6 mg/kg)and madopar(6 mg/kg)were given to rats of the western medicine group by gavage.After 28 days,sucrose solution preference test and open field test were conducted,and the content of serum Ghrelin was detected by ELISA.Results Compared with that of the normal group,the content of Ghrelin in the model group was significantly decreased(P<0.01),while the content of Ghrelin in the western medicine group was not significantly different from that in the model group.The content of Ghrelin in each acupuncture group was significantly increased(P<0.05).Conclusion Acupuncture can obviously improve depression-like behavior of PDD model rats,and can increase the content of Ghrelin in serum.
