黄芪汤对糖尿病肾病小鼠肾脏病变的影响

Effects of Huangqi Decoction(黄芪汤)on Renal Lesion in Diabetic Nephropathy Mice

目的观察黄芪汤对糖尿病肾病db/db小鼠肾脏病变的影响。方法40只6周龄db/db小鼠随机分为模型组,黄芪汤高、中、低剂量组,每组各10只。分别给予0.5%羧甲基纤维素钠和黄芪汤1.08 g/kg,0.36 g/kg,0.12 g/kg灌胃,1次/d。15只6周龄的db/m小鼠随机分为db/m对照组(7只)和db/m黄芪汤高剂量组(8只)分别给予0.5%羧甲基纤维素钠和1.08 g/kg黄芪汤灌胃,1次/d。每4周检测1次各组小鼠的体质量、血糖、血压、尿量、饮水量、摄食量,连续灌胃14周后,20周龄时进行生化指标、肾组织病理学检测。结果与db/db模型组比较,黄芪汤可剂量依赖性的降低db/db小鼠的体质量、尿量、饮水量、摄食量,降低血糖、收缩压、血清糖化蛋白浓度、胰岛素浓度、胰岛素抵抗指数,改善葡萄糖耐量和胰岛素耐量,改善肾功能和肾脏病理情况,降低尿蛋白,提高肾小球的滤过率。结论黄芪汤可以明显改善db/db小鼠肾脏损害,具有延缓糖尿病肾病病情发展的作用。

Objective To observe the protective effect of the Huangqi Decoction(黄芪汤)on db/db mice diabetic nephropathy.Methods Forty mice(6-week-old,db/db)were randomly assigned into model group,Huangqi Decoction of high,medium and low dose groups with 10 in each group.Animals in the model group were intragastrically given 0.5%sodium carboxymethyl cellulose and mice in Huangqi Decoction groups were administered Huangqi Decoction with different doses(1.08 g/kg,0.36 g/kg,0.12 g/kg)once a day.Fifteen mice(6-week-old,db/m)were rando mLy divided into control group(n=7)and Huangqi Decoction high dose group(n=8)which were given 0.5%sodium carboxymethyl cellulose and Huangqi Decoction(1.08 g/kg)once a day respectively.Body weight,blood glucose,blood pressure,urine volume,water intake,food intake of mice in each group were examined every four weeks.After fourteen weeks of intragastric administration,biochemical indicators and renal histopathology of mice(20-week age)were tested.Results Compared with those of the model group,Huangqi Decoction concentration-dependently decreased the body weight,urine volume,water intake and food intake among db/db mice(P<0.01 or P<0.05),improved glucose tolerance and insulin resistance(P<0.01 or P<0.05),lowered blood glucose,systolic blood pressure,serum glycosylated hemoglobin concentration,insulin concentration and insulin resistance index.Renal functions and renal pathological conditions were improved.Urine proteins were reduced.GFR in nephridial tissues were increased.Conclusion Huangqi Decoction shows significant renal protective effect on diabetic nephropathy mice.It can improve renal function and reduce proteinuria.
