中药对中剂量电离辐射致胎盘氧化损伤的影响

摘要 目的观察补肾解毒方对核辐射孕鼠胎盘抗氧化能力的影响。方法将60只C57BL/6J孕鼠随机分为空白对照组、辐射模型组、补肾解毒方组,每组20只,补肾解毒方组给予补肾解毒方13.5 g/(kg·d)灌胃,其他两组给予0.2 mL生理盐水灌胃,各组均灌胃21 d。辐射模型组、补肾解毒方组两组小鼠于灌胃第11天接受一次性4Gy 60Coγ-射线的全身照射。于辐射后第1天、第10天各取10只小鼠剖腹取胎盘组织检测胎盘组织超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)和丙二醛(MDA)含量。结果MDA含量:辐射后第10天,辐射组与空白组比较,孕鼠胎盘MDA含量明显升高(P<0.01),中药组与辐射组及辐射后1天比较孕鼠胎盘MDA含量下降有统计学意义(P<0.01);SOD活性(U/L):辐射后第10天,辐射组、中药组与空白组比较,孕鼠胎盘SOD活性差异有统计学意义(P<0.05),中药组与辐射组比较孕鼠胎盘SOD活性差异显著(P<0.01);中药组辐射后10天较辐射后1天孕鼠胎盘SOD活性差异无统计学意义(P>0.05);GSH-Px活性:辐射后第10天,辐射组、中药组与空白组比较,孕鼠胎盘GSH-Px活性差异有显著统计学意义(P<0.01),中药组与辐射组比较孕鼠胎盘GSH-Px活性差异有统计学意义(P<0.05);中药组辐射后10天较辐射后1天孕鼠胎盘GSH-Px活性升高有统计学意义(P<0.05)。结论补肾解毒方能改善胎盘组织电离辐射所致氧化损伤。 Objective To observe the effect of Bushen Jiedu Prescription(补肾解毒方)on the antioxidant capacity of placenta in nuclear radiation pregnant rats.Methods Sixty C57 BL/6 J pregnant mice were randomly divided into blank control group,radiation model group and traditional Chinese medicine treatment group,with 20 rats in each group.The traditional Chinese medicine treatment group was given Bushen Jiedu Prescription 13.5 g/(kg·d)by gavage,and the other two groups were given 0.2 mL normal saline by gavage.All groups were given intragastric administration for 21 days.Mice in radiation model group and traditional Chinese medicine treatment group received whole body irradiation of 4 Gy 60Coγ-ray on the 11 th day after intragastric administration.On the 1 st and 10 th day after irradiation,the placental tissues of 10 mice were taken out to detect the contents of superoxide dismutase(SOD),glutathione peroxidase(GSH-Px)and malondialdehyde(MDA)in placenta tissue.Results MDA content:on the 10 th day after radiation,the content of MDA in placenta of pregnant rats in the radiation group was significantly higher than that in the blank group(P<0.01).The content of MDA in placentas of pregnant rats in the traditional Chinese medicine group was significantly lower than that in the radiation group and 1 day after radiation(P<0.01).As for the SOD activity on the 10 th day after radiation,there was significant difference in the placental SOD activity among the radiation group,the traditional Chinese medicine group and the blank group(P<0.05),and the difference between the traditional Chinese medicine group and the radiation group was significant(P<0.01).There was no significant difference in the placental SOD activity of pregnant rats in the traditional Chinese medicine group 10 days after radiation compared with 1 day after radiation(P>0.05).As for the GSH-Px activity on the 10 th day after radiation,there was significant difference in GSH-Px activity among the radiation group,traditional Chinese medicine group and the blank group(P<0.01),and the difference between the traditional Chinese medicine group and the radiation group was statistically significant(P<0.05).The activity of GSH-Px in the placenta of the traditional Chinese medicine group was significantly higher than that of the radiation group on the 10 th day after radiation(P<0.05).Conclusion Bushen Jiedu Prescription can improve the oxidative damage of placental tissue induced by ionizing radiation.
