芪蛭真武汤改善单侧输尿管结扎模型大鼠肾间质纤维化的实验研究 被引量:1

Experimental Study on Improvement of Renal Interstitial Fibrosis in UUO Model Rats with Qizhi Zhenwu Decoction(芪蛭真武汤)

摘要 目的研究芪蛭真武汤对单侧输尿管结扎(UUO)大鼠肾间质纤维化的影响。方法将90只雄性SD大鼠,随机分为假手术组、模型组、芪蛭真武汤高剂量组、芪蛭真武汤低剂量组、厄贝沙坦组,每组18只。模型组及各治疗组行左侧输尿管结扎,假手术组不结扎输尿管,仅分离输尿管后关闭腹腔。术后1 d开始灌胃给药,假手术组、模型组予无菌蒸馏水10 mL·kg^(-1),中药高剂量组予芪蛭真武汤方22.2 g·kg^(-1),低剂量组予芪蛭真武汤方11.1 g·kg^(-1),厄贝沙坦组予厄贝沙坦片20 mg·kg^(-1)。各组大鼠分别于术后7,14,21 d取材,收集血清用以检测肌酐(Scr)、尿素氮(BUN);取结扎侧肾脏制备石蜡切片,HE染色观察组织病理形态,MASSON染色检测纤维化程度,免疫组织化学法检测α平滑肌动蛋白、α-SMAⅠ型胶原蛋白、Col-Ⅰ和FN的表达。结果与假手术组比较,模型组Scr、BUN、胶原容积分数(CVF)及α-SMA、Col-Ⅰ、FN的表达均增高(P<0.01;P<0.05);与模型组比较,中药高剂量组和厄贝沙坦组Scr、BUN、CVF值均较低(P<0.05),各治疗组α-SMA、Col-Ⅰ、FN的表达均较少(P<0.05)。结论芪蛭真武汤能够保护UUO模型大鼠肾脏功能,改善肾脏病理损伤,延缓肾间质纤维化的发生和发展。 Objective To study the effect of Qizhi Zhenwu Decoction(芪蛭真武汤)on renal interstitial fibrosis in UUO model rats.Methods A total of 90 male SD rats were randomly divided into sham operation group,model group,Qizhi Zhenwu Decoction high,middle and low dose groups,and irbecartan group(n=18).The left ureter was ligated in the model group and each treatment group,while the ureter was not ligated in the sham operation group,but the ureter was separated and the abdominal cavity was closed.One day after the operation,the sham group and the model group were given sterile distilled water with the dose of 10 mL·kg^(-1).Qizhi Zhenwu Decoction low dose group was given Qizhi Zhenwu Decoction with the dose of 11.1 g·kg^(-1).The irbesartan group was given Irbesartan Tablet with the dose of 20 mg·kg^(-1).Samples were collected 7,14 and 21 days after surgery,and the serum was collected for detection of creatinine(Scr)and urea nitrogen(BUN).The kidney in ligation side was taken out for the preparation of paraffin section,HE staining was used to observe the histopathologic morphology and MASSON staining was used for detecting fibrosis degree.The immune histochemical method was used to detect the expressions ofα-SMA,Col-Ⅰand FN.Results Compared with those of the sham operation group,the levels of Scr and BUN and collagen volume fraction(CVF)in the model group were all increased,and the expressions ofα-SMA,Col-Ⅰand FN in the model group were increased(P<0.01;P<0.05).Compared with those of the model group,levels of Scr,BUN and CVF were lower in Qizhi Zhenwu Decoction high does group and irbesartan group(P<0.05).The expressions ofα-SMA,Col-Ⅰand FN in all treatment groups were lower(P<0.05).Conclusion Qizhi Zhenwu Decoction can protect renal function of UUO model rats,improve renal pathological damage,and delay the occurrence and development of renal interstitial fibrosis.
