通便汤从肺论治便秘对慢性传输型便秘大鼠肺肠SP、VIP含量的影响 被引量:12

Influence of Tongbian Decoction(通便汤)on Contents of Substance and Vasoactive Intestinal Peptid in Lung Intestinal Tissue of Rats with Slow Transit Constipation

摘要 目的观察通便汤对慢性传输型便秘(STC)大鼠肺肠组织神经递质P物质(SP)、血管活性肠肽(VIP)含量的影响,阐明其治疗便秘从肺论治的可能机制。方法选取40只健康SD大鼠,随机选取10只作为正常组,剩余30只用复方苯已哌啶诱导STC便秘大鼠模型,造模时间为120 d。造模结束后,造模组30只大鼠随机分为空白对照组、治疗药物组(通便汤)、阳性对照组(福松),每组10只大鼠。正常组和空白对照组给予同体积生理盐水,连续给药30 d。记录大鼠体质量变化、24 h大便数量及含水量,测定小肠碳末推进率,观察肺肠组织病理变化,用ELISA测定大鼠肺肠组织SP、VIP含量变化。结果药物干预后,治疗组各组大鼠24 h排便量及大便含水量较模型组明显增加(P<0.01),通便汤在改善大鼠排便方面优于福松,差异有统计学意义(P<0.01);通便汤组碳末推进率与造模组比较差异有显著统计学意义(P<0.01),福松组与造模组比较差异有统计学意义(P<0.05),而通便汤组在推进小肠推进率方面优于福松组(P<0.05);药物干预后通便汤组及福松组大鼠肺肠组织病理切片细胞形态学改变得到修复,通便汤较福松组修复程度高;通便汤及福松干预后与模型组相比肺肠组织SP含量均增加(P<0.05);治疗后福松组及通便汤组大鼠肺肠VIP含量与模型组相比均明显降低(P<0.01),通便汤和福松干预后两组肺肠SP、VIP含量变化无统计学意义(P>0.05)。结论通便汤能够明显改善便秘大鼠的排便功能,其治疗便秘从肺论治可能是通过上调肺肠组织SP含量,下调肺肠VIP含量来达到治疗作用。 Objective To observe the effects of Tongbian Decoction(通便汤)on the contents of substance P(SP)and vasoactive intestinal peptid(VIP)in lung intestinal tissue of rats with slow transit constipation and clarify its possible mechanism of treating constipation from lung.Methods Forty healthy SD rates were randomly selected,10 of them as the normal group and the remaining 30 rates as the model group.The model was induced by Compound Phenylethyl Piperidine and the building time was 120 days.After establishing the constipation model,these rates were randomly divided into blank control group,treatment group(Tongbian Decoction)and positive control group(Fusong).Each group had a total of 10 rats.The normal group and the blank control group were given physiological saline volume,continuously for 30 d.The change of rat body weight,defecate number in 24 hours a day and water content were recorded.The intestinal transmission function and the lung intestinal tissue pathological changes were observed.ELISA was used to test the changes of neurotransmitter SP and VIP.Results After drug intervention,24-hour defecation volume and water content of defecation in the treatment group were significantly higher than those in the model group(P<0.01),and Tongbian Decoction was better than Fusong in improving defecation(P<0.01).The carbon advance rate in the treatment group was significantly better than that of the model group(P<0.01)and there was a significant difference between the Fusong group and the model group(P<0.05).After drug intervention,the morphological changes of lung and intestines in the Tongbian Decoction group and the Fusong group were repaired,and the degree of repair in the Tongbian Decoction group was higher than that in the Fusong group.Compared with that of the model group,the SP contents in lung and intestines of the Tongbian Decoction group and the Fusong group were increased(P<0.05).Compared with that of the model group,the VIP contents in lung and intestines of the two groups were decreased significantly after treatment(P<0.01),but the SP and VIP contents in lung and intestines of the two groups did not change significantly after intervention(P>0.05).Conclusion Tongbian Decoction can significantly improve the defecation function of constipation rats.The treatment of constipation from lung may be achieved by up-regulating SP content in lung and intestine and down-regulating VIP content in lung and intestine.
