八味沉香散对缺血再灌注损伤心肌细胞活性氧及钙离子水平的影响 被引量:9

摘要 目的观察八味沉香散对缺血再灌注损伤人心肌细胞的活性氧、钙离子浓度的影响。方法培养人心肌细胞,随机分为9组:正常组、缺血再灌注组(I/R组)、八味沉香散水提物高、中、低剂量组(60、30、15μg/mL)、八味沉香散醇提物高、中、低剂量组(30、15、7.5μg/mL)、曲美他嗪组(60μg/mL)。以体外缺氧缺糖再复氧复糖建立缺血再灌注损伤模型,检测细胞内活性氧、胞浆和线粒体钙离子浓度。结果模型组细胞受到缺血再灌注损伤刺激后,细胞内活性氧、胞浆和线粒体内钙离子浓度明显增高,八味沉香提取物(水提、醇提)较模型组则明显降低(P<0.05,P<0.01)。结论八味沉香散对缺氧再灌注损伤HCM的大量活性氧产生和钙超载具有抑制作用。 Objective To observe the effect of Bawei Chenxiang Powder(八味沉香散)on levels of HCM reactive oxygen and calcium ion in rats with ischemia reperfusion injury.Methods Cultivate HCM cells were randomly divided into 9 groups:normal group,ischemia-reperfusion group(I/R group),Bawei Chenxiang Powder water extraction high,medium and low dose groups(60,30,15μg/mL),Bawei Chenxiang Powder alcohol extraction high,medium and low dose groups(30,15,7.5μg/mL),trimetazidine group(60μg/mL).The ischemia-reperfusion injury model was established by using in vitro hypoxic-deficient glucose reoxygenation and reoxygenation,and the intracellular concentrations of reactive oxygen species,cytoplasm and mitochondrial calcium ions were measured.Results In the model group,after cells were stimulated by ischemia-reperfusion injury,intracellular reactive oxygen species,intracellular and mitochondrial calcium ion concentration were significantly increased,and those of the Bawei Chenxiang Powder extracts groups(water extraction,alcohol extraction)were significantly lower than those in the model group(P<0.05,P<0.01).Conclusion Bawei Chenxiang Powder can inhibit the production of a large number of reactive oxygen species and calcium overload in HCM after hypoxia-reperfusion injury.
