基于网络药理学的麻黄-甘草药对治疗小儿哮喘作用机制 被引量:13

摘要 目的利用网络药理学技术,探索麻黄-甘草药对治疗小儿哮喘的作用机制。方法基于中药系统药理学技术平台(TCMSP)获取麻黄、甘草的化学成分及其作用靶点,UniProt数据库提取作用靶点的基因名称,通过OMIM数据库获取哮喘相关靶点,将疾病靶点与药物靶点进行交集,筛选得到疾病-药物成分共同靶蛋白。利用Cytoscape软件构建化合物-靶点网络,利用DAVID数据库进行GO和KEGG通路富集分析,通过Cytoscape软件构建"中药活性化合物-关键靶标-主要通路"多维网络关系图。结果筛选出115个活性成分,12个作用于小儿哮喘的蛋白靶点;GO富集(P<0.01)提示有24个生物过程条目、3个分子功能条目、3个细胞组分条目,KEGG富集29条通路(P<0.01),关键靶点包括TNFRSF1A、TNF、IKBKG、TP53、NFKBIA、IKBKB等,关键通路包括细胞凋亡、NF-κB信号通路、TNF信号通路、小细胞肺癌、脂肪细胞因子信号通路、RIG-I样受体信号通路、T细胞受体信号通路、Toll样受体信号通路等。结论该研究通过网络药理学初步揭示了麻黄-甘草药对治疗小儿哮喘的多维调控网络,从多角度探索了麻黄-甘草药对治疗小儿哮喘的潜在分子机制及药理作用,为其的临床应用提供了理论依据。 Objective To explore the mechanism of Mahuang-Gancao(ephedra-licorice)in treating children with asthma by using network pharmacology techniques.Methods The Chinese medicine system pharmacology technology platform(TCMSP)was used to obtain the chemical constituents of Mahuang(ephedra)and Gancao(licorice)and their target targets.The UniProt database extracted the gene names of the target,and acquired the asthma-related targets through the OMIM database,and targeted the disease targets and drug targets.The points were intersected and the disease-drug component common target protein was screened.The compound-target network was constructed by Cytoscape software.The DAVID database was used to analyze the enrichment of GO and KEGG pathways.The multi-dimensional network relationship diagram of"Chinese medicine active compound-key target-main pathway"was constructed by Cytoscape software.Results A total of 115 active components were screened and 12 protein targets were applied to children with asthma.GO enrichment(P<0.01)suggested 24 biologic process entries,3 molecular functional entries,and 3 cellular component entries.KEGG analysis had a total of 29 pathways(P<0.01)and key targets included TNFRSF1 A,TNF,IKBKG,TP53,NFKBIA,IKBKB,etc.Key pathways included apoptosis,NF-κB signaling pathway,TNF signaling pathway,small cell lung cancer,Adipocytokine signaling pathway,RIG-I-like receptor signaling pathway,T cell receptor signaling pathway,Toll-like receptor signaling pathway,etc.Conclusion This study preliminarily reveals the multi-dimensional regulatory network of Mahuang-Gancao(ephedra-licorice)in the treatment of asthma in children through network pharmacology.It explores the potential molecular mechanism and pharmacological effects of Mahuang-Gancao(ephedra-licorice)on children with asthma from various angles,and provides the theoretical basis for its clinical application.
