天麻芎苓止眩片对自发性高血压大鼠肾脏保护效应作用机制研究 被引量:3

摘要 目的观察并探讨天麻芎苓止眩片对自发性高血压大鼠肾脏保护效应,初步讨论其具体作用机制。方法40只13周龄SPF级自发性高血压大鼠及10只相同周龄SPF级Wistar大鼠,按照完全随机对照原则,40只13周龄的自发性高血压大鼠分为模型组、(中药)中剂量组、(中药)低剂量组、对照组(本研究采用洛汀新作为阳性对照药物),每组数量均为10只,10只相同周龄Wistar大鼠作为空白对照组,在实验进行过程中进行血压及尿量监测,在实验第8周处死大鼠,抽取血清观察肾功能及炎症相关指标,并取其肾脏组织进行形态学相关观察及研究。结果中药干预组大鼠及对照组大鼠的血压与模型组大鼠对比有显著下降(P<0.05);中药干预组大鼠尿量增加,提示其有利尿作用;中药干预组大鼠、对照组大鼠的肾功能及炎症相关指标及肾脏组织形态学与模型组大鼠对比有显著改善。结论天麻芎苓止眩片可降低自发性高血压大鼠血压,并具有一定的利尿作用,并可明显减少蛋白尿水平以及改善肾功能,同时可明显改善自发性高血压大鼠的肾脏组织纤维化、间质炎细胞浸润、基底膜糖蛋白沉积、足突细胞吞噬及融合,具有显著的肾脏保护效应,可能与天麻芎苓止眩片的抗炎症因子表达作用相关。 Objective To observe and explore the protective effect of Tianma Xiongling Zhixuan Tablet on spontaneously hypertensive rats and discuss its specific mechanism.Methods Forty 13-week-old SPF spontaneously hypertensive rats were randomly divided intomodel group,Chinese medicine middle dose group,Chinese medicine low dose group,control group(Lotintin was used as positive control drug in this study).Each group had 10,and 10 Wistar rats of the same age were used as the blank control group.The blood pressure and urine volume were monitored during the experiment.The rats were sacrificed at the 8 th week of the experiment.The serum was taken out to observe the renal function and inflammation related indicators,and the kidney tissues were taken out for morphological observation and research.Results The blood pressure of the rats in the Chinese medicine intervention groups and the control group were significantly lower than those in the model group(P<0.05).The urine volume of the rats in the Chinese medicine intervention groups increased,suggesting its diuretic effect.The Chinese medicine intervention groups and control group’s renal function and inflammation related indexes and renal histomorphology of were significantly improved compared with those of the model group.Conclusion Tianma Xiongling Zhixuan Tablet can reduce the blood pressure of spontaneously hypertensive rats and has certain diuretic effect,and can significantly reduce proteinuria and improve renal function.It also cansignificantly improve renal tissue fibrosis and interstitial inflammation in spontaneously hypertensive rats.Cell infiltration,basement membrane glycoprotein deposition,phagocytosis and fusion of podocytes have significant renal protective effects,which may be related to the expressions of anti-inflammatory factors in Tianma Xiongling Zhixuan Tablet.
