基于多指标权重分析与正交设计法优选黄芪女贞方菌质提取工艺 被引量:1

摘要 目的优选黄芪女贞方菌质的最佳提取工艺。方法建立高效液相色谱HPLC-ELSD/DAD检测器测定菌质提取物中黄芪甲苷、特女贞苷含量的测定方法;以黄芪甲苷、特女贞苷含量结合干膏得率为评价指标,分别选择提取次数(A)、料液比(B)、提取时间(C)为考察因素,以L9(34)进行正交试验,用多指标试验全概率评分法依据实验结果优选最佳提取工艺。结果本次实验建立了黄芪女贞方菌质中黄芪甲苷、特女贞苷(HPLC)含量的测定方法;以黄芪甲苷含量为评价指标,影响因素(A3>B3>C2);以特女贞苷为评价指标,影响因素(A3>C3>B3),以干膏得率为评价指标(A3>C2>B3)。加权评分优化后确定的最佳提取工艺条件为(A3>C2>B2),即加8倍量的水,提取3次,每次提取1.5 h。结论两种目标化合物含量测定方法灵敏可靠,优选的提取工艺稳定可行,可用于芪菌正气胶囊的进一步制备。 Objective To optimize the extraction process of mycoplasm from Huangqi-Nuzhen(Astragalus-Ligustrum thibeticum).Methods A high performance liquid chromatography(HPLC)-ELSD/DAD detector was established for the determination of astragalosides and specnuezhenide inmycobacterialextract.The extraction times(A),material-liquid ratio(B)and extraction time(C)were selected as the evaluation indexes,and the orthogonal test was carried out with L9(34).According to the experimental results,thepreparation process was optimized by full probability scoring method for multi-index test.Results In this experiment,a method for the determination of astragalosides and specnuezhenide(HPLC)in mycoplasm from Huangqi-Nuzhen(Astragalus-Ligustrum thibeticum)was established.The content of astragalosides was taken as the evaluation index.The influencing factor was A3>B3>C2 and the specnuezhenidewas A3>C3>B3.The dry ointment yield was A3>C2>B3.The optimal extraction conditions determined by the optimization of weighted score were as follows:A3>C2>B2,that is,adding 8 times of water,extracting 3 times for 1.5 hours each time.Conclusion The two methods for the contentdeterminationof the target compounds are sensitive and reliable,and the optimized extraction process is stable and feasible,and can be used for the further preparation of Qijun Zhengqi Capsules.
