解毒胶囊治疗乳腺癌的机制研究 被引量:2

摘要 目的观察解毒胶囊有效成分对乳腺癌相关特异性指标的影响,并分析其作用机制。方法解毒胶囊10×溶液对大鼠进行灌胃7次制备含药血清,以5%、10%、20%浓度对MCF-7细胞加药,并运用MTT法测定细胞增殖与抑制,采用Western blot法检测Bax、Bcl-2、ERα和PR蛋白的表达。结果与Serum组比较,5%浓度含药血清干预下的OD值升高,差异无统计学意义;10%、20%浓度含药血清干预下的OD值均降低,差异有统计学意义(P<0.05)。与Serum组比较,5%含药血清浓度干预下,ERα蛋白表达下降,差异无统计学意义,PR蛋白的表达量降低,有显著性差异(P<0.05);10%、20%含药血清浓度干预下,ERα、PR蛋白表达均明显下降,差异有统计学意义(P<0.05)。与Serum组比较,Bax蛋白表达量明显升高,有显著性差异(P<0.05);Bcl-2蛋白表达量显著下降,差异有统计学意义(P<0.05)。结论解毒胶囊可能是通过诱导病变细胞凋亡、改善性激素紊乱从而达到治疗乳腺癌的目的。 Objective To observe the effect of the effective components of detoxification capsule on the specific indexes of mammary cancer and analyze its mechanism.Methods Detoxification Capsule 1×solution on rats were intragastrically administrated 7 times to prepare the drug-containing serum,with 5%,10%,20%concentrations of the drug in MCF-7 cells.MTT assay was used for the determination of cell proliferation and inhibition.Western blot was used to detect Bax,Bcl-2,ER and PR protein expressions.Results Compared with the serum group,5%drug-containing serum intervention group’s OD value increased and the difference was not statistically significant.The OD values of 10%and 20%drug-containing serum intervention groups were decreased and the difference was statistically significant(P<0.05).Compared with the serum group,5%drug-containing serum intervention group’s ER alpha protein expression decreased and the difference was not statistically significant.The expression of PR proteins decreased and there was significant difference(P<0.05).The 10%and 20%drug-containing serum intervention groups’ER alpha and PR protein expression were significantly decreased and the difference was statistically significant(P<0.05).Compared with the serum group,the expression of Bax protein was significantly increased and there was significant difference(P<0.05).Bcl-2 protein expression was significantly decreased and the difference was statistically significant(P<0.05).Conclusion Detoxification Capsule may improve the sex hormone disorder through the induction of apoptosis in order to achieve the purpose of treatment of mammary cancer.
