铜离子光双交联海藻酸盐水凝胶支架材料的性能研究 被引量:3

摘要 目的:将铜离子载入光交联海藻酸盐水凝胶中,以期改善其机械强度和生物活性等性能。方法:对海藻酸盐进行铜离子光双交联,通过核磁共振仪分析、红外光谱、扫描电镜、溶胀实验以及活死细胞双染试剂检测载入铜离子对光交联海藻酸盐水凝胶结构和性能的影响。结果:光交联基团和铜离子均能成功地接枝到海藻酸盐水凝胶上,且铜离子的载入对支架主要官能团无明显影响。随着载入铜离子浓度的升高,其孔径依次减小、溶胀率逐渐降低,并能促进小鼠胚胎成骨细胞前体细胞(mouse embryo osteoblast precursor cells,MC3T3-E1)在水凝胶表面的黏附和伸展。结论:载入铜离子能提高光交联海藻酸盐水凝胶的机械强度,并促进细胞在水凝胶表面的黏附和伸展。 Objective:To improve the mechanical property and bioactivity of photocrosslinked alginate hydrogel by crosslinking copper ions.Methods:Alginate was successively crosslink ed by photoinitiator and copper ion,~1H NMR,FTIR,SEM,swelling test and Live/Dead Assay were used to examine the effect of copper ions on the structures and properties of photocrosslinked alginate scaffolds.Results:Photocrosslinked groups and copper ions were successfully grafted onto alginate hydrogels.The loading of copper ions has no obvious effect on the main functional groups of the scaffolds,and the increased concentration of copper ions can decrease the pore size and the swelling ratio,but promote the adhesion and extension of mouse embryo osteoblast precursor cells(MC3 T3-E1)on the surface of the hydrogels.Conclusion:Copper Ion can improve the mechanical property of photocrosslinked alginate hydrogel and promote cell adhesion and extension on the surface of the hydrogels.
