摘要
目的:探究杜仲叶提取物对原发性高血压大鼠血管内皮损伤的保护作用。方法:40只雄性SHR大鼠随机分为模型组(DM)、阳性药组(P)、杜仲叶提取物低、中、高剂量组(LEU、MEU、HEU),8只雄性WKY大鼠为正常组(CK)。给药8周后检测各组大鼠血清NO、eNOs、AngII、TNF-α、IL-6含量,HE染色观察血管形态变化,Masson胶原染色观察血管内胶原沉积情况,电镜观察主动脉超微结构变化,Western Blot检测血管RhoA、ROCK1蛋白表达情况。结果:给药8周后,与DM组比较,杜仲叶提取物显著升高血清NO、eNOs含量(P<0.05),显著降低血清AngⅡ、TNF-α、IL-6含量(P<0.05);改善SHR大鼠主动脉血管形态,抑制胶原纤维沉积和细胞器的肿胀变形;显著下调RhoA、ROCK1蛋白和mRNA表达(P<0.05)。结论:杜仲叶提取物对血管内皮损伤具有保护作用。
Objective:To explore the protective effect of Eucommia ulmoides leaf extract on vascular endothelial injury in essential hypertensive rats.Methods:40 male SHR rats were randomly divided into model group(DM),perindopril group(P),low Eucommia ulmoides leaf extract,medium and high dose group(LEU,MEU,HEU),8 male WKY rats were normal Group(CK).The drug was administered 8 weeks of detection of rat vascular active substance NO,eNOS,Ang II and inflammatory factor TNF-,IL-6 content.HE staining to observe vascular morphology change,Masson staining of collagen to observe intravascular collagen deposition,Observe the ultrastructural changes of the aorta with an electron microscope,Western Blot detecting vascular RhoA,ROCK1 protein expression.Results:After 8 weeks of administration,compared with DM group,Eucommia ulmoides leaves extract significantly increased serum NO and eNOs content(P<0.05),and significantly reduced serum AngⅡ,TNF-,IL-6 content(P<0.05);improved the morphology of the aorta in SHR rats and inhibit the deposition of collagen fibers and the swelling and deformation of organelles,To reduce the production of abnormal organelles;and significantly down-regulate the expression of RhoA and ROCK1 protein.Conclusion:Eucommia ulmoides leaf extract has a protective effect on vascular endothelial damage.
