摘要
目的探讨动脉泵入尤瑞克林(HUK)对急性脑梗死大鼠的保护作用及机制。方法将30只大鼠随机分为假手术组(Sham组)、大脑中动脉闭塞(MCAO)组、HUK组,每组各10只。MCAO组建立永久性MCAO大鼠模型。HUK组建立MCAO模型后经动脉泵入HUK。比较各组大鼠神经功能缺损评分、梗死体积、组织病理学变化,以及神经损伤标志物(S-100β、NSE)水平变化。分析HUK对MCAO大鼠SREBP2/DDAH1/ADMA信号通路的影响。结果MCAO组大鼠神经功能缺损评分明显高于Sham组与HUK组,差异有统计学意义(P<0.05)。MCAO组大鼠梗死体积高于HUK组,差异有统计学意义(P<0.05)。MCAO组大鼠海马神经元表现为严重的损伤,HUK组神经元损伤明显逆转。MCAO组大鼠大脑组织神经损伤标志物S-100β和NSE蛋白表达高于Sham组与HUK组,差异有统计学意义(P<0.05)。MCAO组大鼠大脑组织ADMA蛋白表达高于HUK组与Sham组,SREBP2、DDAH1蛋白表达低于HUK组与Sham组,差异有统计学意义(P<0.05)。结论HUK通过SREBP2/DDAH1/ADMA信号通路发挥对大鼠急性脑梗死的功能保护作用。
Objective To explore the protective mechanism of arterial infusion of Ureklin(HUK)on acute cerebral infarction in rats.Methods A total of thirty rats were randomly divided into Sham group,middle cerebral artery occlusion(MCAO)group and HUK group,with 10 rats in each group.Permanent MCAO rat model was established in MCAO group.In the HUK group,the MCAO model was established and HUK was pumped through the artery.The neurological deficit score,infarct volume,histopathological changes,and the levels of nerve injury markers(S-100β,NSE)were compared between groups.Effects of HUK on SREBP2/DDAH1/ADMA signaling pathway in MCAO rats were analyzed.Results The neurological deficit score of rats in MCAO group was significantly higher than that in Sham group and HUK group,and the difference was statistically significant(P<0.05).The infarct volume of MCAO group was higher than that of HUK group,and the difference was statistically significant(P<0.05).The hippocampal neurons of rats in MCAO group showed severe damage,and HUK group significantly reversed the neuron damage.The expressions of S-100βand NSE protein in MCAO group were significantly higher than those in Sham group and HUK group(P<0.05).The expression of ADMA protein in the brain tissue of rats in MCAO group was higher than that in HUK group and Sham group,and the expression of SREBP2 and DDAH1 protein was lower than that in HUK group and Sham group,and the differences were statistically significant(P<0.05).Conclusion HUK has a protective effect on acute cerebral infarction in rats through SREBP2/DDAH1/ADMA signal pathway.
