诱导性低温复苏对失血性休克大鼠肺PGC-1表达影响

摘要 目的探讨诱导性低温复苏对失血性休克大鼠肺PGC-1表达的影响。方法将18只清洁级成年雄性Wistar大鼠随机分为A组(对照)、B组(37℃~38℃常温复苏)及C组(33℃~34℃低温复苏)3组,每组各6只。A组仅进行外科插管操作,不建立失血性休克模型及复苏;B组和C组在建立失血性休克模型后分别在预定温度下进行复苏。失血性休克复苏180 min后检测大鼠肺组织活性氧和三磷酸腺苷(ATP)水平。实时荧光定量聚合酶链式反应和蛋白免疫印迹法检测肺组织PGC-1 mRNA及蛋白表达水平。结果B组、C组肺组织活性氧水平高于A组,且B组高于C组,差异有统计学意义(P<0.05)。B组、C组ATP、PGC-1 mRNA、PGC-1蛋白表达水平低于A组,且B组低于C组,差异有统计学意义(P<0.05)。结论诱导性低温复苏能够上调失血性休克大鼠肺组织PGC-1表达,促进ATP生成,降低活性氧水平。 Objective To investigate the effect of induced hypothermia resuscitation on expression of PGC-1 in rat lung of hemorrhagic shock.Methods A total of eighteen adult male Wistar rats were randomly divided into group A(control),group B(37℃-38℃resuscitation at normal temperature)and group C(33℃-34℃resuscitation at low temperature),with 6 rats in each group.In group A,surgical intubation was performed only,and hemorrhagic shock model and resuscitation were not established.Group B and group C were resuscitated at predetermined temperature after the establishment of hemorrhagic shock model.The levels of reactive oxygen species(ROS)and adenosine triphosphate(ATP)in lung tissues of rats were detected 180 min after resuscitation from hemorrhagic shock.The mRNA and protein expression of PGC-1 in lung tissues were detected by real-time fluorescence quantitative polymerase chain reaction and western blot.Results The level of ROS in lung tissues of group B and group C was higher than that of group A,and group B was higher than that of group C,with statistical significance(P<0.05).The expression levels of ATP,PGC-1 mRNA and PGC-1 protein in group B and group C were lower than those in group A,and group B was lower than that in group C,with statistical significance(P<0.05).Conclusion Induced hypothermia resuscitation can up-regulate the expression of PGC-1 in the ran lung tissues of hemorrgagic shock,promote ATP production and reduce the level of reactive oxygen species.
