合生元制剂对阿尔兹海默病小鼠影响研究 被引量:1

Effects of synbiotics on mice with Alzheimer′s disease

摘要 目的探讨由乳双歧杆菌、嗜酸乳杆菌、低聚木糖制备的合生元制剂对阿尔兹海默病(AD)小鼠的影响。方法选取24只APP/PS1痴呆小鼠和12只C57BL/6J野生小鼠。采用随机分组的方式将APP/PS1痴呆小鼠分成两组,对照组和治疗组各12只;另将12只C57BL/6J小鼠作为空白组。治疗组和对照组小鼠分别灌胃合生元制剂和低聚木糖,每次给药剂量为0.5 ml/只,空白组小鼠灌胃等量的生理盐水,每天3次,持续灌胃3个月。采用Morris水迷宫观察各组小鼠的记忆和空间学习能力,采用免疫组化检测各组小鼠脑内皮质及海马区β-淀粉样蛋白(Aβ)斑块的沉积情况。结果与空白组比较,在实验的第3、4、5天时,对照组逃逸潜伏期显著延长,差异有统计学意义(P<0.05);与对照组比较,在实验的第4、5天时,治疗组逃逸潜伏期显著缩短,差异有统计学意义(P<0.05)。与空白组比较,对照组穿越平台次数减少,目标象限停留时间缩短,差异有统计学意义(P<0.05);与对照组比较,治疗组穿越平台次数增加,目标象限停留时间延长,差异有统计学意义(P<0.05)。与空白组比较,对照组小鼠脑内海马及皮质区Aβ斑块表达增多;与对照组比较,治疗组小鼠脑内海马及皮质区Aβ斑块表达减少。与空白组比较,对照组脑内海马及皮质区平均光密度增加,差异有统计学意义(P<0.05);与对照组比较,治疗组脑内海马及皮质区平均光密度降低,差异有统计学意义(P<0.05)。结论合生元制剂可有效降低APP/PS1痴呆小鼠脑内海马及皮质区Aβ沉积,显著增强小鼠的记忆和空间学习能力,改善认知功能损伤,从而延缓AD进程。 Objective To investigate the effects of synbiotics of bifidobacterium lactis,lactobacillus acidophilus and xylo-oligosaccharide on Alzheimer′s disease(AD)mice.Methods Twenty-four APP/PS1 dementia mice and 12 C57 BL/6 J wild mice were selected.APP/PS1 dementia mice were randomly divided into 2 groups,with 12 in the control group and 12 in the treatment group.Another 12 C57 BL/6 J mice were used as blank group.Mice in the treatment group and control group were intragastric with synbiotics and xylo-oligosaccharide at a dose of 0.5 ml per mouse.Mice in the blank group were intragastric with normal saline,3 times a day,for 3 months.Morris water maze was used to observe the memory and spatial learning ability of mice in each group,and immunohistochemistry was used to detect the deposition of amyloidβ-protein(Aβ)plaques in cortex and hippocampus.Results Compared with the blank group,the escape incubation period of the control group was significantly prolonged on the 3 rd,4 th and 5 th day of the experiment,and the difference was statistically significant(P<0.05).Compared with the control group,the escape incubation period in the treatment group was significantly shortened on the 4 th and 5 th day of the experiment,with statistical significance(P<0.05).Compared with blank group,the number of crossing platform in control group was reduced and the residence time in target quadrant was shortened,with statistical significance(P<0.05).Compared with the control group,the number of crossing platform in the treatment group increased and the residence time in target quadrant prolonged,with statistical significance(P<0.05).Compared with blank group,the expression of Aβplaques in hippocampus and cortex of control group was increased.Compared with the control group,the expression of Aβplaques in the hippocampus and cortex of the treated group was decreased.Compared with blank group,the average optical density of hippocampus and cortex increased in control group,and the difference was statistically significant(P<0.05).Compared with the control group,the average optical density of hippocampus and cortex in the treatment group decreased,and the difference was statistically significant(P<0.05).Conclusion Synbiotics can effectively reduce Aβdeposition in hippocampus and cortex of APP/PS1 dementia mice,significantly enhance the memory and spatial learning ability of mice,improve cognitive impairment,and delay the process of AD.
